氢气生理盐水对大鼠下肢缺血再灌注引起的肺损伤的保护作用

         

摘要

Objective To evaluate the effect of hydrogen-rich saline (HS) in protecting acute lung injury induced by lower limb ischemia reperfusion in rats.Method 30 adult male SD rats were chosen as the subjects and were randomly divided into 5 groups: normal saline control group (group CON), model group (group LIR), hydrogen saline (2.5ml/kg, 5ml /kg, 10ml /kg) preconditioning group (HS1, HS2, HS3 groups).3-hour reperfusion was conducted for the model group rats after a rubber band ligation on the lower limbs had been executed for 3 hours.The rats treated with hydrogen saline 30 minutes before ligation were given intraperitoneal injection of hydrogen saline (2.5ml/kg, 5ml /kg, 10ml /kg).The normal saline group were given the same amount of saline as given to the other two groups.3-h after reperfusion, sample carotid artery blood was taken for blood gas analysis and conducted bronchoalveolar lavage on the side and then tested the concentration of MDA and GSH on the other side of lung by tissue pathology detection.Then detected the concentration of cell factors TNF-alpha and IL-1 beta as collected in the alveolar lavage fluid.Findings The LIR group’s pathological score was significantly higher than that of the CON group while its oxygenation index lower than that of the CON group (P<0.01).The pathological score of HS2 group and HS3 group was lower than that of the LIR group while the two groups’ oxygenation index was higher than that of the LIR group (P <0.01).Compared with the CON group, the LIR group’s concentration of MDA and the inflammation factors TNF-alpha and IL-1 beta in bronchoalveolar lavage fluid significantly increased (P <0.01); GSH concentration in the lung tissue, however, decreased significantly (P <0.01).HS1, HS2 and HS3 groups’concentration indexes of MDA and inflammation factors TNF-alpha and IL-1 beta in bronchoalveolar lavage fluid was significantly higher than those as observed in the LIR group (P <0.01).Concentration of GSH in the lung tissue was also higher than that of the LIR group (P <0.01).Conclusion HS can reduce ALI in the lower limbs caused by LIR.The effect of HS may be correlated with the inflammatory reaction and the oxidative stress reaction.%目的:探讨氢气生理盐水(Hydrogen-rich saline, HS)对大鼠下肢缺血再灌注(Limb ischemia reperfusion, LIR)引起的急性肺损伤(Acute lung injury, ALI)的保护作用。方法成年雄性 SD 大鼠30只,随机均分为5组:生理盐水对照组(CON 组)、模型组(LIR 组)、氢气盐水(2.5 mL/kg、5 mL/kg、10 mL/kg)预处理组(HS1、HS2、HS3组)。LIR 组大鼠使用橡皮筋高位结扎双下肢3小时后恢复灌注3 h;氢气盐水处理组大鼠在结扎前30分钟腹腔注射不同剂量氢气盐水,其余两组分别静脉给予相同容量的生理盐水。恢复灌注3小时后取大鼠颈内动脉血进行血气分析,并进行一侧肺泡灌洗,ELSIA 法检测另一侧肺组织中 MDA 和 GSH 的浓度。检测收集的肺泡灌洗液中 TNF-α和IL-1β的浓度。结果 LIR 组病理学评分显著高于 CON 组而氧合指数低于 CON 组;HS2和 HS3组病理学评分低于LIR 组而氧合指数高于 LIR 组,均有统计学意义(P <0.01)。与 CON 组相比,LIR 组大鼠肺泡灌洗液中 TNF-α和IL-1β浓度、肺组织中 MDA 浓度显著升高;肺组织中 GSH 的浓度显著降低(均 P <0.01)。 HS1、HS2和 HS3组肺泡灌洗液中 TNF-α和 IL-1β浓度、肺组织中 GSH 浓度均显著高于 LIR 组,MDA 浓度低于 LIR 组(均 P <0.01)。结论HS 可减轻大鼠下肢 LIR 引起的 ALI,HS 的作用可能与其减轻炎症反应和氧化应激反应有关。
