Objective To study the relationship between protein-rich tyrosine kinase 2( PYK2 ) and left ventricular hypertrophy( LVH ) in rats with renavascular hypertension, and to explore the mechanism in LVH results from renovascular hypertension. Methods Twenty male SD rats were divided randomly into two groups: operation group and sham-operation group. There were 10 rats in each group. The two kidney one clip( 2K1C ) operation was performed to establish the left ventricular hypertrophy model with renovascular hypertension in operation group. Sham-operation group underwent sham operation. The blood pressure of rats was measured before operation,at the 4th,8th, 12th week after operation, respectively. The rats were killed 12 weeks after operation, whose hearts were taken out for detection of the corresponding indices. Results ① Blood pressure in operation group was more than 150 mmHg 4 weeks after operation. Compared with sham-operation group,blood pressures in operation group at the 4th,8th, 12th week after operation increased significantly(P < 0. 05 ). ② Left ventricular mass index( LVM ), left ventricular wall thickness( LVWT ), interventricular septal thickness( IVST ) in operation group were significantly larger than those in sham-operation group( P < 0. 05 ), while left ventricular dimension( LVD ) in operation group was smaller than that in sham-operation group( P < 0.05 ). ③ Protein expression of PYK2 in operation group was significantly higher than that in sham-operation group( P < 0.05 ). Conclusion LVH results from renovascular hypertension might be closely associated with the increased protein expression of PYK2.%目的 探讨富脯氨酸蛋白酪氨酸激酶(PYK2)与肾血管性高血压大鼠左心室肥厚的关系及其发病机制.方法 20只雄性SD大鼠,随机分成手术组及假手术组,每组各10只.手术组行二肾一夹手术建立肾血管性高血压左室肥厚模型,假手术组行假手术.分别于术前及术后4周、8周、12周检测鼠尾动脉血压1次;术后12周处死动物,取心脏检测相应指标.结果 (1)术后4周时,手术组大鼠收缩血压大于150 mmHg,与假手术组大鼠相比,手术组大鼠术后4周、8周、12周时血压均明显升高(P＜0.05).(2)手术组大鼠左心室质量指数(LVMI)、左心室壁厚度(LVWT)、室间隔厚度(IVST)均明显大于假手术组(P＜0.05),而左心室腔内直径(LVD)小于假手术组(P＜0.05).(3)手术组大鼠心肌组织PYK2蛋白表达明显高于假手术组(P＜0.05).结论 PYK2蛋白表达增加与大鼠肾血管性高血压左心室肥厚密切相关.
