高海拔地区缺氧预处理对大鼠脑缺血再灌注损伤模型脑梗塞体积的影响

         

摘要

Objective:To explore the effect of hypoxic preconditioning on the volume of cerebral infarction in the model of ischemia-reperfusion in adult wistar rats in high altitude. Methods: 60 male adult Wistar rats were randomly divided into hypoxic preconditioning group and control group, 30 rats in each group. In hypoxic preconditioning group, animals were put in hypoxia cabin for 3 hours a day for three days which mimic 5500m altitude, then these rats were fed in Xining city (the altitude of Xining is about 2200m). In control group, rats were fed in Xining normally. Three days after hypoxic preconditioning, these rats were prepared for middle cerebral artery occlusion-reperfusion model. After operation, brain tissue was taken out to undergo TTC staining; the volume of cerebral infarction was calculated and compared. Results: The volume of cerebral infarction was 35.07 ±2.69% in hypoxic preconditioning group, and it was 34.75 ±3.79% in control group. There was no significance difference in the volume of cerebral infarction between the two groups. Conclusion: Hypoxic preconditioning(mimic 5500m altitude, 3 hours a days for 3 days) could not reduce the volume of cerebral infarction in Wistar rats in high altitude.%目的: 探讨高海拔地区缺氧预处理对成年Wistar大鼠脑缺血再灌注损伤模型脑梗塞体积的影响. 方法:60只雄性Wistar大鼠,随机分为缺氧预处理组及对照组,每组30只,缺氧预处理组将大鼠置低压氧舱(模拟5500米海拔高度)进行缺氧预处理,每日处理3h,连续处理3d,随后置西宁地区(海拔2200米)正常饲养;对照组置西宁地区正常饲养.缺氧预处理后第3d,成功制备右侧大脑中动脉脑缺血/再灌注损伤模型. 术后取脑组织行TTC染色, 对脑梗塞体积进行比较. 结果: 缺氧预处理组脑梗塞体积为(35.07±2.69)%,对照组脑梗塞体积为(34.75±3.79)%,经两独立样本t检验,两组脑梗塞体积无显著性差异. 结论:缺氧预处理(5500米,每天3h,连续3d)对高海拔地区成年Wistar大鼠脑缺血再灌注模型脑梗塞体积无显著保护作用.
