树莓品种'波拉纳'组培繁殖体系建立

         

摘要

为了探索树莓品种"波拉纳"组织培养的最佳培养条件,进行工厂化育苗,大量繁殖优良苗木,以"波拉纳"茎段为试材,采用正交试验设计法,对外植体消毒方式、基本培养基种类、试管苗的增殖培养方法及生根培养方法等环节进行了研究.结果表明:茎段的最佳灭菌处理为75%酒精30 s+0.1%升汞8 min+2%次氯酸钠2 min,污染率、褐化率和成活率分别为2.88%、15.93%和81.11%.茎段在WPM、1/2 MS、MS三种培养基上均能萌动、展叶.但在MS培养基上,腋芽嫩绿、生长健壮,因此,初代培养最佳培养基为MS+6 BA0.5 mg/L+NAA0.1 mg/L.试管苗最适增殖培养基和植物生长调节剂组合为MS+KT1.0 mg/L+NAA0.1 mg/L+6-BA0.5 mg/L,增殖倍数为5.14,苗高为2.89 cm.最佳生根培养基及植物生长调节剂浓度为MS+IBA0.5 mg/L,25 d生根率达到100%,生根数达到6.14条,主根粗壮.%In order to explore the optimal tissue culture conditions for Raspberry (Rubus idaeus) Cultivar "Polana", which can produce a large scale of high quality seedlings by factory nusery, stem segments were selected as experiment materials, studying the sterilization methods of the explant, basal medium types, proliferation methods and rooting method by orthogonal test. The results showed that the optimal sterilization method of stem segments was 75% Ethanol for 30 s +0.1%HgCl2for 8 min + 2% NaClO for 2 min. The contamination percentage, browning percentage and survival percentage were 2.88%, 15.93% and 81.11% respectively. Stem segments could be germinated and expanded leaf on three kinds of media (WPM、1/2 MS、MS). But on the MS medium, the axillary bud was bright green and vigorous. Thus, the optimal primary culture medium was MS+6-BA0.5 mg·L-1+NAA0.1 mg·L-1. The optimal proliferation medium and hormone combination of plantlet was MS+KT1.0 mg·L-1+NAA0.1 mg·L-1+6-BA0.5 mg·L-1, multiplication index was 5.14 and the height of plantlet was 2.89 cm. On the 25thday, rooting percentage was 100%, the rooting number was up to 6.14 and the main root were strong.
