[Objective] To investigate the role of total saponins from Momordica charantia Descourt (TSMC) on the mRNA level of proinsulin in type 2 diabetic rats.[Method] Wistar rats were randomly divided into diabetic model group,diabetic model treated with TSMC [300ng/ (kg · d)],TSMC [100mg/ (kg · d)] and normal control group.[Result] The mRNA level ofproinsulin in rats pancreas tissue was examined by semi-quatitiativa RT-PCR.Compared with model control group,the mRNA expression of proinsulin in TSMC groups was higher than that of diabetic model group (P < 0.01),and the levels of FBG and FINS decressed.[Conclusion] TSMC could promote proinsulin gene transcripts,stimulate insulin biosynthesis and secretion and decrease blood glucose.%目的:探讨苦瓜总皂苷对2型糖尿病大鼠血糖及胰岛素原基因(proinsulin)mRNA水平的影响.方法:将Wistar大鼠随机分为4组,A:正常对照组;B:模型组;C:苦瓜总皂苷高剂量组;D:苦瓜总皂苷低剂量组.8周后,测定空腹血糖、胰岛素;以半定量RT-PCR法检测胰岛素原基因转录水平的表达.结果:与模型组比较,苦瓜总皂苷组空腹血糖、胰岛素降低,苦瓜总皂苷组大鼠胰岛素原基因mRNA水平显著增高(P<0.01).结论:苦瓜总皂苷促进2型糖尿病大鼠胰岛素原基因转录,迅速升高血胰岛素以降低血糖.
