辽宁省脊髓灰质炎实验室细胞敏感性实验方法的建立与应用

摘要

Objective To prepare the quality control standard strain (QC) of Liaoning Provincial polio laboratory(LNPPL), to evaluate the sensitivity of the cell line to the polio virus in LNPPL, and to provide reliable quality assurance for the routine monitoring of the polio laboratory. Methods 96-microwells plate'S culture and titration was used. Results In the Qc of LNPPL,the fluctuation of titre was±0..5 log 10 CCID 50 in 3 independent cellular sensitivity tests.At the sametime, China Sabin Test Reference Strain(CSTRS)provided by the National Polio Laboratory in viral disease control and preventionstation in the National CDC was used as parallel contrast.The fluctuation of titre were also±0.5 log 10 CCID 50 in 3 independent cellularsensitivity test comparing with its own standard provided by CSTRS.The results were also within the normal range of 9 conventional cell line sensitivity test. Conclusion The result of Qc in LNPPL was in accordance withexperimental requirement.And the sensitivity of the cell line to poliovirus used in LNPPL was not dropping,which was sensitive andefective.%目的：制备辽宁省疾病预防控制中心脊髓灰质炎(脊灰)实验室质量控制标准毒株(QC),评价辽宁省脊灰实验室所用细胞系对脊灰病毒的敏感性,为脊灰实验室常规监测工作提供可靠质量保证。方法采用96孔微量培养板滴定法。结果辽宁省脊灰实验室2013年制备的QC 3次独立的细胞敏感性实验结果的滴度波动在±0.5 log 10 CCID 50以内，同时用中国疾病预防控制中心病毒病预防控制所国家脊灰实验室提供的已知滴度的Sabin参考株(CSTRS)做平行对照，CSTRS原始株3次滴度结果与其本身提供的参考值相比较，其滴度波动均在±0.5 log 10 CCID 50范围内。开展的9次常规细胞系敏感性实验，结果也均在正常范围内。结论辽宁省脊灰实验室QC结果符合实验要求，辽宁省脊灰实验室所用细胞系对脊灰病毒的敏感性未下降,是敏感、有效的。
