氢气饱和生理盐水对高糖所致内皮细胞内活性氧生成和凋亡的影响

             

摘要

Objective To study the effects of hydrogen rich saline on endothelial reactive oxygen species(ROS) induction and apoptosis induced by high glucose. Methods Cells were divided into three groups,including control group,high glucose group,and H2 group (high glucose + hydrogen rich saline). The level of intracellular ROS was detected by FACS. The levels of super oxide dismutase(SOI)) ,and glutathione peroxidase(GSH Px) mRNA were detected by realtime PCR. The apoptosis of ceils was analyzed by cell death ELISA. Results The level of intracellular ROS was reduced in the H2 group,compared with the high glucose group (2.75±0. 37 vs. 6.35±1. 29,P<0.01). The levels of SOD and GSH-Px mRNA were higher in the H2 group than those in the highglucose group (SOD:0.74±0. 06 vs. 0.26±0.03,P<0.01;GSH Px:0. 68± 0.05 vs. 0. 31 ± 0. 04,P<0. 01). The apoptosis of cells in the H2 group was decreased,compared with the high glucose groups (0.38±0. 02 vs. 0.89±0. 05,P<0.01). Conclusion hydrogen rich saline can decrease the high glucose induced intracellular ROS induction, increase the antioxidase, and protect the high glucose induced apoptosis of endothelial cell.%目的 探讨氢气(H2)饱和生理盐水对高糖所致内皮细胞内活性氧(ROS)生成和凋亡的影响.方法 细胞分为对照组、高糖组、高糖+H2饱和生理盐水组(H2组)3组,流式细胞仪检测细胞内ROS,Realtime PCP检测氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)mRNA,细胞凋亡ELISA试剂盒检测细胞凋亡.结果 H2组内皮细胞内ROS较高糖组明显减少(2.75±0.37 vs 6.35±1.29,P<0.01),而SOD和GSH-Px mRNA水平明显增高(SOD:0.74±0.06 vs 0.26±0.03,P<0.01;GSH-Px:0.68±0.05 vs 0.31±0.04,P<0.01),细胞凋亡水平明显降低(0.38±0.02 vs 0.89±0.05,P<0.01).结论 H2饱和生理盐水可减少高糖诱导的内皮细胞内ROS增多和抗氧化酶的消耗,并减少高糖诱导的细胞凋亡.
