不同时间应用重组人血管内皮抑制素对小鼠肺腺癌SPC-A1生长的影响

         

摘要

目的 观察在不同时间应用重组人血管内皮抑制素(恩度)后肺腺癌SPC-A1肿瘤组织的生长情况,为重组人血管内皮抑制素的临床应用提供理论依据.方法 将24只小鼠按随机分组方法,分成4组(Ⅰ组:接种肿瘤日前6d开始给药;Ⅱ组:接种肿瘤当日开始给药;Ⅲ组:接种肿瘤日后第6天开始给药;Ⅳ组:对照组),每组6只,且各组给予的重组人血管内皮抑制素剂量均为3 mg·kgJ-1·d-1;通过对各实验组与对照组小鼠体内肺腺癌SPC-A1肿瘤体积大小及肿瘤微血管密度的比较,观察在不同时间应用重组人血管内皮抑制素后对肿瘤血管生成的影响.结果 在开始实验18d后,Ⅰ组肿瘤体积为(2078±185)mm3,肿瘤微血管密度为(42±4);Ⅱ组肿瘤体积为(928±126)mm3,肿瘤微血管密度为(36±5);Ⅲ组肿瘤体积为(2512±603) mm3,肿瘤微血管密度为(41±5);Ⅳ组肿瘤体积为(4009±780)mm3,肿瘤微血管密度为(47±6);Ⅱ组肿瘤体积和微血管密度与对照组(Ⅳ组)相比均显著下降,差异有统计学意义(P<005),而Ⅰ组和Ⅲ组分别与Ⅳ组相比肿瘤体积和微血管密度差异均无统计学意义.结论 重组人血管内皮抑制素具有抗肿瘤血管生成作用,最佳给药时间是接种肿瘤当天.%Objective To observe the effects of rh-endostatin (Endu) on mice lung adenocarcinoma SPC-A1 tumor group at different time, and to provide the theoretical basis for the clinical application of Endu. Methods 24 mice were randomized into 4 groups (I group given drugs at 6 d before tumor inoculation; Ⅱ group given drugs at the day of tumor inoculation; Ⅲ group given drugs at 6 d after tumor inoculation; Ⅳ group assigned as control group) (n=6 each). Moreover, the Endu doses given to every group were all 3 mg·kg-1·d-1. The size of in vivo mice lung adenocarcinoma SPC-A1 tumor and intratumoral microvessel density (MVD) was then compared in experimental groups and control group. And the effects of rh-endostatin on mice lung adenocarcinoma SPC-A1 tumor group at different time were observed. Results At 18d after experiment, the size of tumor and MVD were (2078±l85)mm3 vs (36±5), (2512±603)mm3 vs (36±5), (2512±603)mm3 vs (41±5) and (4009±780)mm3 vs (47±6) in I , Ⅱ ,Ⅲ and Ⅳ groups, respectively. Notably, the size of tumor and MVD in Ⅱ group were significantly decreased as compared with those in control group (P<0.05), but there was no difference in the size of tumor and MVD between 1 group and IV group, and between Ⅲ group and IV group. Conclusion Endu had an anliangiogenesis function, and the most suitable time for drug use was the day of inoculation.
