2-Methoxy-6-acetyl-7-methyljuglone exhibits anti-inflammatory properties via MKP-1 in LPS-stimulated RAW264.7 macrophages

Aim Recent study have reported 2-Methoxy-6-acetyl-7-methyljuglone (MAM) for its potential antimicro- bial, neuroprotective and anticancer activity. However, the anti-inflammation effects of MAM remains to be elucida- ted. We investigated the anti-inflammation activity of MAM. Methods RAW 264.7 macrophages were exposed to LPS with or without MAM. Inducible nitric oxide synthase (iNOS) expression and signaling molecules activated by LPS were evaluated. Results LPS-induced iNOS expression and nitric oxide (NO) expression was suppressed by MAM. MAM attenuated p38MAPK in cells treated with LPS. In addition, MAM caused an increase in MKP-1 expres- sion, which could suppress p38MAPK phosphorylation. Conclusions MAM may activate MKP-1, which then de- phosphorylates p38MAPK, resulting in iNOS down-regulation in LPS-stimulated RAW264.7 macrophages. The pres- ent study indicate that MAM may possess the potential to alleviate LPS-associated inflammatory disorders.
