Aim To observe the influence of Buyanghuanwutang( BYHWT ) on the DNA-binding activity of cAMP response element binding protein ( CREB ) and CCAAT/enhancer binding protein( C/EBP ) in hippocampus of vascular dementia ( VD ) rats. Methods The rats were randomly divided into sham-operation group, VD model group, BYHWT-treated group and nimodipine-treated group. VD rat models were prepared by four-vessel occlusion , and physiological saline solution . BYHWT and nimodipine were administered respectively to four groups by gavage. Electrophoretic Mobility Shift Assay ( EMSA ) was used to observe the change of DNA-binding activity of CREB and C/EBP.Results The DNA-binding activity of CREB of rat hippocampus was significantly lower in VD model group than that in Sham-operation group( P < 0. 01 ). Compared with VD model group, both nimodipine-treated group and BYHWT-treated group were definitely higher ( P < 0. 01 ). There was no significant difference between BYHWT-treated group and nimodipine-treated group( P > 0. 05 ). Conclusion BYHWT can improve the DNA-binding activity of CREB and C/EBP in hippocampus of VD rat.%目的 观察补阳还五汤对血管性痴呆大鼠海马CREB(cAMP反应元件结合蛋白)、C/EBP(CCAAT/增强子结合蛋白)DNA结合活性的影响.方法 实验分为假手术组、模型组、补阳还五汤组和尼莫地平组.采用改良的4血管法制备血管性痴呆模型,分别给予生理盐水、补阳还五汤、尼莫地平灌胃干预,用凝胶电泳迁移率改变分析法(EMSA)测定各组大鼠CREB和C/EBP的DNA结合活性.结果 与假手术组比较,模型组大鼠海马组织CREB的DNA结合活性明显降低(P0.05).结论 补阳还五汤可提高VD大鼠海马组织 CREB、C/EBP与DNA的结合活性.
