基于肺与大肠相表里的'肠病及肺'动物模型制备

             

摘要

目的:制备适用于研究"肺与大肠相表里"理论的"肠病及肺"大鼠模型.方法:20只SPF级雄性Wistar大鼠,随机分为模型组和假手术组,模型组采用腹腔注射人工胃液后2h再注射大肠杆菌的方法制备的"肠病及肺"大鼠模型,造模后6h测定大鼠肛温,取全段结肠中的粪便用于结肠粪便含水量测定,取末端回肠和肺组织用于髓过氧化物酶(MPO)活性和病理学检测.结果:与假手术组相比,模型组大鼠体温升高,结肠粪便含水量减少,回肠和肺组织MPO活性增加(1.39±0.21 vs 0.72±0.16和1.51±0.26 vs 0.86±0.20,P<0.05);同时模型组大鼠显微镜下可见回肠绒毛排列紊乱、萎缩或脱落,肺间质水肿、出血,部分肺泡塌陷、融合;透射电镜下可见肠上皮细胞间紧密连接破坏,线粒体嵴断裂,肺泡Ⅱ型上皮细胞胞浆浓缩,细胞核固缩,伴空泡化.结论:采用腹腔注射人工胃液后注射大肠杆菌的方法制备的"肠病及肺"大鼠模型,原发病在腹腔,同时具备肠和肺损伤,是研究"肺与大肠相表里"脏腑相关理论的一种合适的实验动物模型.%Objective To prepare a model of"Large Intestine Disease Causing Pulmonary Disease"in rats which suitable for research the theory of"Exterior and Interior Relationship between Lung and Large Intestine". Methods SPF male Wistar rats (n=20) were randomly divided into sham-operation group and model group. The model of"Large Intestine Disease Causing Pulmonary Disease"was prepared by intraperitoneal injection of artificial gastric juice and Escherichia coli. At 6 h after modeling, the rectal temperature and the water content in the whole colon fecal were measured; the MPO activity and the histopathological changes of ileum and lung were examination. Results Compared with sham operation group, the body temperature of rats in model group was increased, the water content of colon fecal was decreased, and the MPO activity of ileum and lung was in?creased(1.39 ± 0.21 vs 0.72 ± 0.16 and 1.51 ± 0.26 vs 0.86 ± 0.20,P<0.05). Microscopically, the ileal villi of the model group were disorganized, atrophic or detached, the alveolar septa became markedly damaged and the fu?sion of alveoli appeared. Transmission electron microscopy showed that the tight junctions between intestinal epi?thelial cells were disrupted, the mitochondrial cristae were broken. In alveolar type II epithelial cell of the model group, cytosolic concentration, pyknosis, and vacuolization were observed. Conclusion The rat model of"Large Intestine Disease Causing Pulmonary Disease"is a suitable model to research the theory of"Exterior and Interior Relationship between Lung and Large Intestine".
