mGluR1在单眼形觉剥夺性弱视大鼠外侧膝状体的表达变化

     

摘要

目的 观察视觉发育敏感期内单眼形觉剥夺性弱视大鼠外侧膝状体mGluR1蛋白的表达变化规律,探讨其在弱视发病机制中的作用.方法 2周龄大鼠随机分为正常对照组和实验组,缝合实验组大鼠单侧眼睑30天,建立剥夺性弱视模型.免疫组化技术检测mGluR1蛋白的表达情况.结果 免疫组化结果:剥夺眼对侧外侧膝状体组较剥夺眼同侧外侧膝状体组及正常对照组mGluR1蛋白的表达明显减少,差异有显著性(P>0.05),而正常对照组与剥夺眼同侧外侧膝状体组相比,差异无显著性(P>0.05).结论 视觉发育敏感期内,单眼形觉剥夺后外侧膝状体mGluR1蛋白的表达降低,导致神经元之间的突触联系减少,致突触可塑性发生变化可能是弱视发生的机制之一.%Objective To observe the regulation of expression of mGluR1 protein at lateral geniculate body (LGB) of monocular form-deprivation amblyopia rat within sensitve period of visual development, in order to explore its role in the pathogenesis of amblyopia. Methods Rats aged 2 weeks old were randomly divided into normal control group and experimental group, and unilateral eyelid suture was performed on experimental group rats for 30 days and the models of form -deprivation amblyopia were established. We used immunohistochemistry technology to detect the changes of mGluR1 protein expression. Results Compared with normal control group and ipsilateral LGB of deprived eyes group, the expression of mGluR1 protein in contralateral LGB of deprived eyes group are remarkable decreased,and the dif-ferences were significant (P >0.05). There were no significant differences between normal control group and ipsilateral LGB of deprived eyes group ( P>0.05). Conclusions After form-deprivation,the expression of mGluR1 protein is de-pressed in the sensitve period of visual development, which induce the connection of synapses are reduced between neu-rons,then synaptic plasticity changed, and this may be one of the pathogenesis of amblyopia.
