水通道蛋白AQP9在帕金森病脑组织中的表达及作用

         

摘要

Objective To observe the expression changes of aquapor‐9 ( AQP9) and its role in rat brain tissues of Parkin‐son’s disease (PD).Methods Parkinson's disease model in rats was made. The expression changes of AQP9 was detected by HE stain ,immunohistochemistry method and RT‐PCR.Results The detection of immunohistochemistry and RT‐PCR showed that the expressions of AQP9 and AQP9 mRNA presented dynamic changing process in Parkinson's disease group. The expres‐sions of AQP9 and AQP9 mRNA in the positions of left substantia nigra pars compacta (SNC) and the ventral tegmental area (VTA) in Parkinson's disease group were higher than that in the sham operation group at each time point after operation (ex‐cept for at 2‐week time point). Conclusion The high expression of AQP9 possibly plays an important role in the pathogenesis of Parkinson's disease by regulation of water electrolyte and energy metabolism.%目的:观察大鼠帕金森病(Parkinson’disease ,PD)水通道蛋白9(aquaporin 9,AQP9)在脑组织中的表达变化,研究AQP9在帕金森病脑组织发病机制中的作用。方法制造帕金森病动物模型,应用HE染色、免疫组织化学、RT‐PCR等方法观察大鼠脑组织AQP9表达变化。结果免疫组织化学和RT‐PCR检测结果显示,帕金森病组脑组织 AQP9和AQP9 mRNA 的表达呈动态变化过程,帕金森病组大鼠术后各时间点(除外术后2周时间点)在鼠左侧黑质致密部(SNC )及中脑腹侧被盖区(VTA)区AQP9和AQP9 mRNA表达均比假手术组高。结论高表达的AQP9,可能通过调节水电解质与能量代谢,从而在帕金森病发病机制中起重要作用。
