大鼠脑室出血后脑干蓝斑内去甲肾上腺素能神经元表达变化的研究

         

摘要

目的:观察大鼠脑室出血后脑干蓝斑(L C )内去甲肾上腺素能神经元的表达变化,探讨其在脑室出血中的损伤作用,对诊断疾病、预测病情及预后有实际的临床意义。方法将60只大鼠随机分为正常组、假手术组、脑室出血组(IV H组)。IV H组取自体动脉血方法制备PIV H模型,假手术组大鼠只切开头皮和颅骨钻孔,不做自体动脉血注射。分别在术后5个时间点取脑干蓝斑染色,并计数DBH阳性细胞数。结果 IVH组蓝斑内出现大量深染的DBH阳性神经元细胞,形态与正常组、假手术组相似,染色加深,阳性细胞数较正常组、假手术组在6h、1d、3d、5d、7d显著增加,差异有统计学意义(P<0.05)。结论蓝斑是神经系统去甲肾上腺素(NE)的最重要来源,同时50%的去甲肾上腺素能神经元分布在蓝斑,IVH后,蓝斑内DB H阳性神经元表达增加,脑干组织去甲肾上腺素含量增加,去甲肾上腺素在脑室出血后脑组织损伤中可能起重要作用,并对外周组织产生继发性损害,导致病情加重。%Objective To observe the changes of DBH positive neuron of brainstem locus coeruleus (LC) of the rat after ventricular hemorrhage to research the damage mechanism in the ventricular hemorrhage;thus it has practical clinical meaning for diagnosing disease ,predicting illness and instructing the treat after recovery. Methods 60 experimental animas were grouped into normal group ,shame operation group and IVH group randomly ;the IVH model prepared by the method of injec‐ting their own blood for group IVH ;only opening the scalp and drilling on the skull for the shame operation group ,taking the locus coeruleus of the normal group ,shame operation group and IVH group to chromo after operation at five time points and counted DHB positive neurons. Results There were much deeply chromo DBH positive neuron in the locus coeruleus of IVH group with similar form to the normal group and shame operation;the number of positive cells and chromo were obviously in‐creased compared with the normal group and shame operation (P<0.05). Conclusion Locus coeruleus is the nervous system of norepinephrine (NE) is the most important source ,at the same time ,50% of norepinephrine neurons in locus coeruleus ,af‐ter IVH ,the expressing of the DBH in the locus coeruleus of the rat brain stem is improved ,the brain tissue nordrenalin level is increased ,thus NE might has important effect on the brain tissue damage after the ventricular hemorrhage ,and causes other organs secondary lesion ,and aggravates illness.
