天山地区红景天属植物根中红景天苷及苷元酪醇的定量分析

         

摘要

目的:建立高效液相色谱方法测定天山地区红景天属植物根中红景天苷及苷元酪醇的含量.方法:液相色谱分离系统采用岛津VP5.0溶剂输送系统,LC10A TVP自动进样器,PDA检测器,色谱柱VP-ODS 4 μm,150 mm×4.5 mm,流动相为甲醇-乙腈-水(25:5:70),柱温40 ℃,分析时间15 min,检测波长275 nm,进样量20 μL.结果:方法简单、灵敏和精确.2物质的线性范围分别为:0.171 4~2.228 mg*mL-1(r=0.998 5)和0.201 2~2.616 mg*mL-1(r=0.999 6),回归方程分别为Y=1.885×104+5.247×105X 和Y=-1.284×104+7.077×105X .结论:发现Rhodiola rosea L.根中含有(44.38±0.7)mg*g-1红景天苷和(16.37±0.9)mg*g-1苷元酪醇,并生长于相对较低海拔高度,认为是一个更具开发利用前景的目标植物.%Objective:To establish a HPLC method to determine the contents of salidroside and tyrosol in roots of Rhodiola L.plants grown at Tianshan mountain.Method:The HPLC system consisted of Shimadzu Class VP5.0 multi-solvent delivery system;Shimadzu LC 10A TVP auto sampler; Shimadzu LC 10A TVP photodiode array detector(PDA),Shimadzu temperature control module.Column:Shimadzu VP-ODS 4 μm,150 mm×4.5 mm.Solvent:isocratic with methanol,acetonitrile and water(25∶ 5∶ 70).Flow rate:0.5 mL*min-1at 40 ℃.Analysis time:15 min.Detection:at 275 nm with injection volume 20 μL.Results:The methods were simple,sensitive and accurate.The linearity of the method was observed in the ranges from 0.171 4 to 2.228 mg*mL-1and 0.201 2 to 2.616 mg*mL-1;regression equations were Y=1.885×104+5.247×105X(r=0.998 5),and Y=-1.284×104+7.077×105X(r=0.999 6),respectively.Conclusion:It is found that Rhodiola rosea L.,containing(44.38±0.7)mg*g-1salidroside and(16.37±0.9)mg*g-1tyrosol and growing at lower altitude,is considered as the target plant for further research.
