视黄酸信号在不同波长色光照射后豚鼠视网膜中的研究

摘要

Objective Retinoic acid (RA) is the only extrinsic biochemical candidate that could act as a growth controller,the present study was designed to evaluate the retinoic acid signals in retina of guinea pig eyes with different monochromatic illumination.Methods Ninety 2-week-old guinea pigs were divided into 3 groups randomly and were raised under green (530nm) ( n =30),blue (430nm) ( n =30 ) or white lighting(5000K) (n =30) conditions.The axial length was measured with Cinescan A/B ultrasonography before,2 weeks,4 weeks,8 weeks during experiment.After the biometrical measurement finished,9 guinea pigs were sacrificed randomly in each group and retina was dissected.The RA level in retina was detected by High Performance Liquid Chromatography ( HPLC).The expressions of retinoic acid-binding proteins Ⅰ (CRABP- Ⅰ ) and retinoie acid receptor-β( RAR-β) protein and mRNA were assayed by Western-blot and Real-time PCR,respectively.The experiment and use of animals followed the Standard of Association for Research in Vision and Ophthalmology.Results 1.The axial length among the three goups showed a significant difference at 4 weeks during experiment ( F =3.946 ; P < 0.05 ),the green lighting group showed the fastest growing,the axial length of the three groups are ( 8.36 ± 0.11 ) mm (green),(7.88 + 0.49 ) mm( blue),7.98±0.32 mm( white) respectively at 8 weeks during experiment; 2.The RA levels demonstrated rna same trend with the axial length. at 8 weeks during experiment the RA levels are 4.846±0.56 μg/grn(green).3.583 ±0.26 μg/g ( blue)、4.419±0.36μg/g ( white) respectively; 3.Changes of the rnexpression of CRABP- I and RAR-β occurred at different time8,a significant difference of RAR-β among rnthree groups appeared earlier,the green lighting group showed a highest leveL Conclusion The different rnlevels of retinoic acid signals were showed in retina of guinea pig eyes under different monochromatic rnillumination The eye growth was significantly faster in green group than in blue group and white group,rnmeanwhile,the RA level,the expressions of CRABP- I and RAR-p in the retina were higher in green group rnthan the other two proups. These findings suggest that RA may act as a messenger in eye growth promoted by rndifferent monochromatic illumination.%目的 视黄酸(RA)是目前所知的惟一可以通过外源性途径导致眼轴增长的物质,本研究旨在对不同波长色光照射后豚鼠视网膜中视黄酸转运信号转运系统进行研究.方法 出生2周英国短毛豚鼠90只,随机分为绿光组(波长530 nm) (n =30)、蓝光组(波长430 nm)(n=30)以及白 光(色温5000K)对照组(n=30).入组前、3个组内色光照射2周、4周及8周后,所有豚鼠用Cinescan A/B超仪测定眼轴长,完成生物学测量后随机选取9只处死并取视网膜,用高效液相色谱仪( HPLC)法测量视网膜中的RA水平,以免疫印迹法定量测定视网膜中视黄酸结合蛋白-Ⅰ(CRABPI)和视黄酸核受体β(RAR-β)的蛋白含量,并用Real-time PCR法测量视网膜中CRABP-Ⅰ和RAR-β的mRNA水平,不同组间指标差异采用ANOVA单因素方差分析,组间采用LSD两两分析.结果 3个组间眼轴长度自干预4周起出现明显差异(F=3.946;P<0.05),其中绿光组眼轴生长最快,干预8周后分别为绿光组(8.36±0.11)mm、蓝光组(7.88 +0.49)mm、白光组(7.98±0.32) mm;3个组间RA水平自干预4周起出现统计学意义(F=4.382;P<0.05),其中绿光组RA水平明显高于蓝光组,干预8周时差异更加明显,分别为绿光组(4.846±0.56) μg/g、蓝光组(3.583±0.26) μg/g、白光组(4.419±0.36) μg/g;3个组间CRABP-Ⅰ的mRNA及蛋白水平自干预8周后差异均出现统计学意义(F=4.984,P<0.05;F=5.312,P<0.05),而3个组间RAR-β的蛋白水平干预2周后mRNA水平即出现统计学意义(F=4.4.47;P<0.05),干预4周后蛋白水平出现明显差异(F=4.568;P<0.05),绿光组的CRABP-Ⅰ和RAR-β明显高于蓝光组.结论 不同波长色光照射后豚鼠眼轴长度及视网膜视黄酸信号转运系统呈现出不同水平,绿光组眼轴增长最快,且视网膜RA、CRABP-Ⅰ和RAR-β水平均明显高于蓝光组以及白光组,视网膜视黄酸信号在不同波长色光照射引起的豚鼠眼球生长发育中可能起到重要的调控作用.
