活体和离体脑血管痉挛动物模型的比较

摘要

Objective To investigate different characteristics between in vivo and in vitro cerebral vasospasms model. Methods Using trans verteral artery angiography, cerebral vasospasm were observed by injecting platelet plasma and intact red cells into canine chiasmatic cister in 22 dogs. Other 13 dog were killed by femoral arterial exsanguinatian, then the basilar arteries were put off, made helical strip and place in Loker-Ring solution, contraction of serotonin (5-HT) were examined using an isometric tension recording system at intact endothelium and endothelium removed group.Results In vivo significant cerebral vasopasm was induced by platelet plasm at half hour and novasospam at 4, 72 hours. Intact red cells produced vasopasm little at half hour, mild vasospasm at 4 hours, severe vasospam at 72 hours. In vitro, 1×10-9M of 5-HT produced arterial strip contraction, 1×10-7M produced significant contraction. Contraction induced by each isodimer was augmented in the endotheliam-removed strips. Conclusions In vivo model appreciate to observe vasospasm for long time and to observe arterial pathological change. In vitro mode appreciate single factor test and entdothelium-removed.%目的 探讨离体和活体脑血管痉挛动物模型的优缺点。方法 对通过22条犬椎动脉造影、视交叉穿刺注入血小板、红细胞,不同时间观察基底动脉收缩情况。13条犬麻醉后,股动脉放血、猝死、摘出基底动脉作成动脉条,Loker-Ring营养液中,观察5-HT作用后收缩情况,动脉条一组作了内皮剥脱。结果 注入血小板血浆,注射当时引起稍重度收缩,注射后4小时、72小时无明显收缩。注入红细胞,注射当时无明显收缩,注射4小时轻度痉挛,72小时重度痉挛。离体动脉条5-HT 1×10-9M即引起收缩,1×10-7M达到高峰。内皮去除组收缩强于内皮完整组。结论 活体脑血管痉挛动物模型,适合长时间观察收缩情况及病理改变。离体实验模型最适合单因素试验及内皮去除。
