脑膜瘤血小板源生长因子及受体表达与脑膜瘤增殖活性及凋亡研究

摘要

Objective To explore the role of PDGF in the development of meningiomas.Methods The expression of PDGF and its receptor were detected in 61 human meningiomas and 7 fetal arachnoid tissues by immunohistochemistry. The proliferative activity and cell apoptosis of meningiomas were evaluated by the proliferating cell nuclear antigen labeling index(PCNA LI) and terminal deoxynucleitidyl transferase mediated dUTP nickend labeling (TUNEL) method, respectively.Results The results showed that only two fetal arachnoid tissues expressed PDGF β receptor. None of the fetal arachnoid tissues expressed PDGFBB, PDGFAA and PDGF α receptor. Most of the 61 meningiomas expressed PDGFBB and PDGF β receptor, and the immunostaining intensity were higher in atypical meningiomas than that in benign types. There was no significant difference between the different types of benign meningiomas. The average PCNA LI and apoptosis labeling index(%) were higher in atypical meningiomas than that in benign types, and there was no significant difference between the different types of benign meningiomas. The expression of PDGFBB and PDGF β receptor was significant enhanced in ascending order from low PCNA LI meningiomas to high ones. Conclusions The overexpression of PDGFBB/PDGFR β in meningomas was correlated with grade of miningiomas and the proliferative activity of meningiomas; PDGF-BB/R β autocrine loop may play an critical role in the pathogenesis of meningiomas.%目的 探讨血小板源生长因子(PDG-F)在脑膜瘤发生和发展中的作用。方法 采用免疫组化检测新鲜脑膜瘤组织中PDGF及其受体表达,用TUNEL法检测细胞凋亡情况,同时进行细胞增殖核抗原(PCNA)检测。结果 脑膜瘤高表达PDGF及其受体,且以表达PDGFB链及PDGF β受体为主,部分脑膜瘤表达水平很低的PDGFA链,几乎不表达PDGFα受体;非典型性脑膜瘤PDGFBB、PDGFR β蛋白表达水平及PCNA指数高于良性脑膜瘤,但同时其凋亡也增加。结论 PDGFBB/R β自分泌环可能在脑膜瘤的发生和发展过程中起着重要作用。
