大鼠跟腱异位骨化模型中低氧诱导因子-1α及血管内皮生长因子的表达及作用

     

摘要

Objective To observe expression changes of hypoxia inducible factor-1α(HIF-1α) and vascular endothelial growth factor (VEGF) in rat Achilles tendon heterotopic ossification model, and to investigate effects of HIF-1αand VEGF signal path in occurrence and progression of heterotopic ossification. Methods A total of 60 SD rats were randomly divided into experimental group and control group, and each group contained 30 cases. Heterotopic ossification model was made in the experimental group. Achilles tendon tissues were taken in 5 and 10 weeks for detection of HIF-1αand VEGF expression levels by RT-PCR and Western-blot test. Results In 5 and 10 weeks after operation, the control group had obviously lower HIF-1αand VEGF expression levels than the experimental group (P<0.05). Conclusion Remarkably increased HIF-1αand VEGF expression levels in early stage of rat Achilles tendon heterotopic ossification provides important effects for occurrence and progression of heterotopic ossification.%目的:通过观测大鼠异位骨化模型中低氧诱导因子-1α( hypoxia inducible factor-1α, HIF-1α)及血管内皮生长因子(vascular endothelial growth factor, VEGF)表达变化,探讨HIF-1α、VEGF信号通路在异位骨化发生发展中的作用。方法 SD大鼠60只随机分为实验组和对照组,各30只,实验组制备异位骨化模型。于5、10周取跟腱组织, RT-PCR及免疫印迹试验(Westen-blot)检测两组局部组织中HIF-1α及VEGF表达水平。结果术后5、10周对照组HIF-1α及VEGF表达量均显著低于实验组(P<0.05)。结论在大鼠跟腱异位骨化早期, HIF-1α及VEGF表达明显升高,提示其对异位骨化发生、发展有重要作用。
