目的 研究乳酸脱氢酶-A亚基(LDH-A)在人胶质瘤中的表达与意义.方法 收集67例脑胶质瘤标本(WHO Ⅱ级25例、Ⅲ级22例、Ⅳ级20例)及10例行内减压术切除的正常脑组织标本,采用免疫组织化学方法研究LDH-A在不同病理级别胶质瘤及正常脑组织中的表达情况.结果 在正常脑组织中未见阳性表达,LDH-A阳性表达率在Ⅲ、Ⅳ级胶质瘤中分别为68.2%、75.0%,均显著高于Ⅱ级胶质瘤(32.0%)(P<0.05).胶质瘤病理级别与LDH-A蛋白表达强度呈正相关(r=0.522,P<0.05).结论 LDH-A在胶质瘤中高度表达,其阳性表达率及表达强度随着病理级别增加而上升,这提示胶质瘤中存在糖酵解通路的活化.%Objective To investigate the expression and its significance of lactate dehydrogenase-A subunit (LDH-A) in human gliomas. Methods Sixty-seven glioma samples were obtained from 25 patients with WHO grade II gliomas, 22 grade Ⅲ ones and 20 grade IV ones, and 10 normal brain tissue samples were taken from patients undergoing internal decompression surgery. The expressions of LDH-A in gliomas and normal brain tissues were detected by immunohistochemistry. Results LDH-A positive rates of gliomas grade Ⅲ and grade IV were 68.2% and 75.0% respectively, which were significantly higher than that of grade II (32.0%) (P < 0.05). No positive expression of LDH-A was observed in the normal brain tissues. The pathological grade was positively correlated with protein expression level of LDH-A (r = 0.522 ,P< 0.05). Conclusions LDH-A is highly expressed in human gliomas, its positive expression rate and the expression level are increased with increase of glioma grade, suggesting that glycolytic pathway may be activated in glioma.
