目的:探讨牛乳铁蛋白对大鼠空肠上皮细胞株IEC-6的细胞活性、增殖能力的影响。方法体外培养IEC-6细胞,MTT法检测不同浓度牛乳铁蛋白对IEC-6细胞的活性,Brdu标记检测牛乳铁蛋白对细胞增殖能力的影响,流式细胞术检测细胞周期。结果牛乳铁蛋白促进IEC-6细胞活性,且呈浓度依赖效应,牛乳铁蛋白处理浓度为100µg/ml时,IEC-6细胞活性显著上调(P<0.05)。牛乳铁蛋白可通过调节细胞周期促进IEC-6细胞增殖,牛乳铁蛋白在100µg/ml时细胞Brdu增殖活性发生显著性上调(P<0.05),在200µg/ml牛乳铁蛋白处理浓度时增殖能力达到顶峰。牛乳铁蛋白可通过调控IEC-6的细胞周期,从G1期向S期转化。结论牛乳铁蛋白可能通过调控细胞周期从G1期向S期转化,从而促进大鼠空肠上皮细胞株IEC-6细胞活性及增殖能力。%ObjectiveTo study the influence of bovine lactoferrin on the cell viability and cell proliferation, and the protective effects in IEC-6 cells treated by lipopolysaccharides(LPS).MethodThe rat jejunum epithelial cell lines IEC-6 were purchased. The beneficial effects on cell viability and cell proliferation in IEC-6 of bovine lactoferrin were detected by MTT assay and Brdu assay separately. The cell cycle distribution was examined by flow cytometry. The inflammatory effects were induced by LPS in IEC-6 and the cytokines were evaluated by real-time PCR and ELISA test.ResultBovine lactoferrin significantly increased the cell viability of IEC-6 and promoted IEC-6 proliferation via regulating the cell S stage in the whole cell cycle. Bovine lactoferrin could down regulate the release of cytokines in IEC-6 induced by LPS. ConclusionBovine lactoferrin could promote the cell viability and cell proliferation, and show protective effects in IEC-6 cells.
