目的:建立HPLC法测定地锦草中游离没食子酸和总没食子酸的含量.方法:采用Agela Odyssil C1s (4.6 mm×250 mm,5 μm)色谱柱;流动相为甲醇-0.5%磷酸溶液(8:92);检测波长270 nm;流速1.0 mL· min-1;柱温30℃.结果:没食子酸在0.034 0~1.700 2 μg范围内呈良好线性关系(r=1.000 0),游离没食子酸和总没食子酸平均回收率分别为99.63%和99.34%.结论:该方法操作简便准确,灵敏度高,重复性好,可用于地锦草中没食子酸的质量控制.%Objective:To establish a method for quantitative determination of free gallic acid and total gallic acid in Euphorbiae Humifusae Herba by HPLC.Methods:The HPLC procedure was performed on the chromatographic column of Agela Odyssil C1s (4.6 mm × 250mm,5μm) at 30℃,the mobile phase was methanol-0.5% phosphoric acid solution(8:92) with the flow velocity of 1.0 mL · min-1,and the detection wavelength was 270 nm.Results:Gallic acid showed a good linear relationship with peak area at a range from 0.0340 μg to 1.7002 μg(r =1.0000),the free gallic acid recovery was 99.63% and the total gallic acid recovery was 99.34%.Conclusion:The method is simple,accurate with high sensitivity and good repeatability,and can be used for the quality control of gallic acid in Euphorbiae Humifusae Herba.
