Objective To explore the influence of PLA-Poly-PTMC with GDNF on the protein expression of GAP-43 in spinal cord injury of rats.Methods To explore the protein expression of GAP-43 at molecular biological level with Western blot to make certain the difference between PLA-PTMC/ GDNF and others.Results B,C,D groups showed high protein expression at 2 weeks,and there is no significant difference among these groups.D group had sig-nificant higher expression at 4 weeks that that of two weeks.C,D groups showed high protein expression at 6 weeks than other groups and all groups showed higher expression.At 8 weeks,all groups showed lower expression of GAP-43,but D group showed more expression than others.Conclusion Conclusion The functional Recovery in the composite catheter group (Group D)function is best,the repair of the recovery of Spinal Cord Injury is superior to a single appli-cation of PLA-PTMC catheter group or a single application of GDNF group.%目的探讨聚乳酸-聚三亚甲基碳酸酯/GDN 导管对大鼠脊髓损伤区 GAP-43蛋白表达的影响。方法通过 Western Blot 法从分子生物水平对 GAP-43蛋白的表达进行分析,从基因水平上来检测植入 PLA-PTMC/ GDNF复合导管与其它治疗组 GAP-43基因表达上的差异。结果术后第2周时,B 组、C 组和 D 组 GAP-43蛋白表达比正常组增高;B 组、C 组和 D 组各组间 GAP-43蛋白的表达水平无显著性差异。术后第4周时,D 组的 GAP-43蛋白表达与2周时明显增高;术后第6周时,各组 GAP-43蛋白表达仍增加,但增幅不同,C 组和 D 组与正常对照组、A 组、B 组相比明显增高,D 组与 C 组相比无显著性差异;8周时,各组 GA-P43蛋白的表达与6周相比均有所均下降,D 组与正常对照组及 A 组、B 组和 C 组相比较仍增高。结论PLA-PTMC/ GDNF 复合导管较单一应用 PLA-PTMC 或 GD-NF 更有利于轴突再生进而促进脊髓损伤的功能恢复。
