眩晕定方对后循环缺血性眩晕家兔血脂水平及椎动脉粥样硬化的影响

         

摘要

Objective To investigate the effect of Xuanyunding prescription(XYDP) on the blood lipids and vertebrarterial atherosclerosis in rabbits with posterior circulation ischemic vertigo(PCIV). Methods Sixty rabbits were randomly divided into normal control group with 8 cases and intervener group with 52 cases,among which 8 randomly selected as XYDP prophylaxis group. The medium - dose XYDP was administered by gavage to XYDP prophylaxis group from the end of fourth week of the model establishing process to the termination of the experiment. Out of the remaining 44 cases,32 rabbits were consistent with the standard model. They were then randomly divided into 4 groups:Model control group,XYDP medium - dose group,XYDP high - dose group and Yangxue Qingnao granule group,with 8 cases in each group. Except for the normal control group,the models of the other groups were established according to the literature concerned. The rabbits in normal control group were fed with normal feedstuff. The remaining groups were modeled with controlled high lipid feedstuff and establish for 6 weeks consecutively. Drug or double distilled water was administered for 2 weeks by gavage after the models were essentially established. The levels of total cholesterol (TO, triglyceride (TG) and low- density lipoprotein cholesterol (LDL - C) and blood rheology of the rabbits in all groups were measured after the treatment,and pathology change on the arteria vertebralis was observed. Results Compared with the model control group, the levels of TC,TG,LDL - C and blood rheology in XYDP medium - dose group,XYDP high - dose group,XYDP prophylaxis group and Yangxue Qingnao granule group, were significantly lowered (P<0. 05 or P<0. 01). The levels of TC,TG,LDL — C and blood rheology in XYDP high - dose group and XYDP prophylaxis group were decreased markedly compared to those in XYDP medium - dose group and Yangxue Qingnao granule group(P<0. 05 or P<0. 01). The thickness of vertebral artery intima in XYDP high - dose group and XYDP prophylaxis group was mildly incrassated. The arteries mildly stenosis or normal in this two groups. Conclusion XYDP could reduce blood lipids and decrease blood rheology. XYDP could inhibit vertebrarterial atherosclerotic formation and development.%目的 探讨眩晕定方对后循环缺血性眩晕(PCIV)家兔模型血脂水平及椎动脉粥样硬化的影响.方法 从60只家兔中随机选取8只设为空白对照组,其余52只采用文献方法造模.造模动物中,随机选取8只为眩晕定预防组,在造模进行至第4周末,给予眩晕定中剂量干预直至实验结束;其余44只实际造模成功32只,随机分为模型对照组、养血清脑颗粒组及眩晕定高、中剂量组,每组各8只.空白组予自由饮水,进食普通家兔饲料;造模组自由进水、限量普通饲料+限量高脂饲料喂食.造模总计6周,造模成功后分别给予药物干预2周.实验结束后抽血检测血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)和三酰甘油(TG);测定全血黏度,并观察椎动脉内膜形态学改变.结果 与模型组相比,眩晕定高剂量组、眩晕定中剂量组、眩晕定预防组、养血清脑颗粒组血清TC、TG、LDL-C水平及血黏度显著降低(P<0.05或P<0.01),且眩晕定高剂量组及预防组对血脂水平及血黏度的改善优于眩晕定中剂量组与养血清脑颗粒组;眩晕定高剂量组及预防组动脉内膜轻度增厚,管腔无狭窄或轻度狭窄.结论 眩晕定方可有效降低家兔血脂水平,改善椎动脉内膜粥样硬化.
