蝉花菌丝对单侧输尿管结扎大鼠肾间质uPA、PAI-1蛋白及mRNA表达的影响

         

摘要

Objective: To study the effect of Cordyceps Sobolifera Mycelium ( CSM ) on renal tubulointerstitial fibrosis through observing the expression of urokinase - type plasminogen activator ( uPA ) and plasminogen activator inhibitor - 1 ( PAI - 1 ) in rat with obstructive nephropathy. Methods: The rat with interstitial fibrosis induced by unilateral ureter obstruction ( UUO ) was researched for 25 days. The experimental models were divided into normal control group, sham operation group, UUO group, Cordyceps sinensis ( CS ) -treated group, high dose CSM ( HDCSM ) - treated group, middle dose CSM ( MDCSM )- treated group and low dose CSM ( LDCSM ) - treated group. The protein and mRNA expression of uPA and PAI- 1 was examined by immunological histochemis-try and in situ hybridization technology. Results: The protein and mRNA expression of PAI - 1 was significantly up - regulated, although the expression of uPA did not change markedly, in UUO group compared with normal control group and sham operation group ( P < 0. 05 ~ 0. 01 ). Treatments with CS and CSM down - regulated the expression of PAI - 1 ( P < 0.05 ~ 0.01 ) and up - regulated the expression of uPA (P<0.05~0.01 ). And the expression of PAI - 1 was lower in HDCSM - treated group than that in CS - treated group or LDCSM -treated group ( P<0.05 ~0. 01 ). Conclusion: Our study suggests that CSM may ameliorate renal tubulointerstitial fibrosis induced by unilateral ureter obstruction in rat models,which is probably attributable to altering the disturbance of PAI -1 and uPA expression.%目的:探讨蝉花菌丝对单侧输尿管结扎大鼠肾间质uPA、PAI-1蛋白及mRNA表达的影响.方法:采用单侧输尿管结扎的方法制备单侧输尿管梗阻(UUO)肾间质纤维化大鼠模型.实验分组为正常组、模型组、假手术组、冬虫夏草组、蝉花高剂量组、蝉花中剂量组和蝉花低剂量组,实验周期为25 d.采用免疫组织化学和原位杂交方法,分别检测肾组织uPA、PAI-1蛋白及其mRNA表达,使用图像分析系统对结果进行处理,采用SPSS11.5统计软件统计分析.结果:模型组肾小管间质中PAI-1蛋白及mRNA的表达均明显高于正常组和假手术组(P<0.01);而uPA蛋白及mRNA的表达则与正常组和假手术组差异无统计学意义(P>0.05);虫草组和不同剂量的蝉花菌丝组的PAI-1蛋白及mRNA表达量明显低于模型组(P<0.05~0.01);而uPA蛋白及mRNA的表达量则明显高于模型组(P<0.05~0.01);蝉花菌丝大剂量组的PAI-1蛋白及mRNA的表达量又较蝉花小剂量组和虫草组为低(P<0.05~0.01).说明蝉花菌丝能下调UUO大鼠肾小管间质PAI-1蛋白及mRNA的高表达,上调UUO大鼠uPA蛋白及mRNA的低表达,并呈明显的量效依赖关系.结论:本研究表明蝉花菌丝通过调节UUO大鼠肾小管间质uPA/PAI-1蛋白及mRNA的表达紊乱而发挥抗肾间质纤维化的药理作用.
