Objective: To investigate the effect of SVC-Ⅲ on the expression of NF-κB in THP1 cells. Methods: The expression of p65 and IKK1 mRNA ia THP1 cell was measured with RT-PCR. The expression of p65 protein was detected by Western blot. Meanwhile, the activity of transcription factor NF-κB was detected with method of luciferase reporter gene. Results: 1 μg/L SVC- Ⅲ could promote the p65 expression at both mRNA and protein level,enhanced the activity of transcription factor NF-κB. However, there was no effect at concentration of 10 μg/L and there was an inhabitory effect at concentration of 100 μg/L on NF-κB activity. Conclusion: SVC- Ⅲ could regulate the expression and activity of transcription factor NF-κB in dose-dependent ways in THP1 cells.%目的:研究蝎素组分Ⅲ(Scorpion venom crude Ⅲ,SVC-Ⅲ)对 THP1细胞中NF-κB转录因子的影响.方法:用RT-PCR方法检测THP1细胞中p65和IKK1基因的表达;Western blot检测细胞p65蛋白的表达;荧光素酶报告基因的方法观察NF-κB的转录活性.结果:1 ìg/L浓度的SVC-Ⅲ在mRNA和蛋白水平均对NF-κB有促进表达作用,并增强NF-κB的转录活性;10 ìg/L浓度的SVC-Ⅲ则未显示出促进作用;100 ìg/L浓度的SVC-Ⅲ反而表现出一定的抑制作用.结论:SVC-Ⅲ能以剂量依赖的方式调节THP1细胞NF-κB转录因子的表达和活性.
