目的 探讨及总结成功构建大鼠心肌梗死动物模型方法.方法 优化建模操作,结扎左冠状动脉前降支构建大鼠心肌梗死模型,应用利多卡因预防室性心律失常的发生,予心电图及超声心动图等检测手段评估模型.结果 模型组成功率达83%;术后20minⅡ导联心电图ST段明显上抬约0.2mV,提示结扎成功及心梗模型初步建立;术后4周超声心动图显示模型组大鼠心室短轴缩窄率显著下降(P<0.01),射血分数显著下降(P<0.05),而左室收缩末期内径及舒张末期内径明显高于假手术组(P<0.01);心肌组织HE染色镜下观察显示细胞排列紊乱,细胞核固缩甚至碎裂,左心室游离壁心肌组织大量坏死.结论 该心肌梗死动物模型构建过程科学严谨,造模成功率高,结果可靠.%Objective To investigate and summarize the method of successful establishment of myocardial infarction model in rat. Method Optimizing the operations course of ligating left anterior descending coronary artery to make the model of myocardial infarction in rat,we applied lidocaine to prevent ventricular arrhythmias in the surgery and used ECG and echocardiogram to evaluate model after operation.Result The success rate in model group is up to 83%.Twenty minutes after operation,SD rates' ECG showed the ST segment elevated about 0.2mv and it signified the establishment of myocardial infarction model in rat;Four weeks after operation,SD rats' echocardiogram proved that fraction shortening and ejection fraction decreased significantly(P<0.05);The left ventricular end systolic diameter and end diastolic diameter was significantly higher than sham operation group(P <0.01);Observing the histological section under the light microscope,it was found that pathological cardiac muscle cells were arranged in disorder,myofilaments fractured and cell nucleus pyknosis and chip occurred. Conclusion The technique possesses the characteristic of being scientific and rigorous and that with high success rate and reliable result.
