Objective To investigate the ratio of Helios+regulatory T cells in the peripheral blood of young people (youth group), healthy elderly (older group) and elderly patients with lung cancer (lung cancer group). Methods Using Flow cytometry to detect the percentage of CD4+Foxp3+T cells and CD4+Foxp3+Helios+T cells in youth group, older group and lung cancer group and compare whether there are differences between groups of people. Results The proportions of CD4+Foxp3+cells in the CD4+cells in peripheral blood of youth group, older group and lung cancer group were (2.77±0.53)%, (3.82±0.61)%, (6.00±2.40)%, respectively. The older group was significantly higher than the youth group (P<0.01);lung cancer group was significantly higher than the older group (P<0.01). The proportions of CD4+Foxp3+Helios+cells in the CD4+Foxp3+cells in peripheral blood of youth group, older group and lung cancer group were (47.67±4.53)%, (26.38±4.50)%, (16.76±4.13)%, respectively. The older group was significantly lower than youth group (P<0.01); lung cancer group was significantly lower than the older group (P<0.01). The proportions of CD4+Foxp3+Helios+ cells in the CD4+Foxp3+cells of the stageⅠ, stageⅡ, and stageⅢlung cancer in elderly were (18.52±2.99)%, (18.57±3.05)%, (12.13±3.23)%respectively. There was no significant difference between the stageⅠand stageⅡpatients (P>0.05), but stageⅢpatient was significantly lower than stageⅠand stageⅡpatients (P<0.01). Conclusion The proportion of Helios+regulatory T cells in youth group is significantly higher than that in older group and lung cancer group, and that higher in stageⅠandⅡthan that in stageⅢlung cancer group.%目的:观察健康青年人(青年组)、健康老年人(老年组)及老年肺癌患者(肺癌组)外周血中Helios+调节性T细胞分布比例。方法流式细胞技术检测青年组、老年组及肺癌患者外周血中CD4+Foxp3+、CD4+Foxp3+Helios+细胞所占比例,同时比较各组人群之间是否存在差异。结果1.青年组、老年组、肺癌组外周血中CD4+Foxp3+细胞占CD4+T细胞的比例分别为(2.77±0.53)%、(3.82±0.61)%、(6.00±2.40)%,青年组明显低于老年组(P<0.01),老年组明显低于肺癌组(P<0.01)。2.青年组、老年组、肺癌组外周血中CD4+Foxp3+Helios+细胞占CD4+Foxp3+T细胞的比例分别为(47.67±4.53)%、(26.38±4.50)%、(16.76±4.13)%,青年组明显高于老年组(P<0.01),老年组明显高于肺癌组(P<0.01)。3.肺癌组中,Ⅰ期、Ⅱ期、Ⅲ期CD4+Foxp3+Helios+细胞占CD4+Foxp3+T细胞的比例分别为(18.52±2.99)%、(18.57±3.05)%、(12.13±3.23)%,肺癌Ⅰ期与Ⅱ期患者无明显区别,而Ⅲ期则明显降低(P<0.01)。结论Helios+调节性T细胞在青年组比例明显高于老年组及肺癌组,早期肺癌明显高于晚期肺癌。
