预防性电针对不同月龄大鼠骨骼肌形态和腓肠肌中 AKT、mTOR、p70S6K 蛋白表达的影响

     

摘要

Objective To observe the effect of preventive electroacupuncture on the morphology of musculoskeletal system of differ -ent month'age rats and protein synthesis expression of gap -associated protein under the pathway IGF /AKT.Methods 30 SD rats were ran-domly divided into control and electroacupuncture groups , with 15 rats in each group.The control group was divided into 3 subgroup accord-ing to the month'age, namely 8 /14 /16 month; the electroacupuncture group was divided into 3 subgroup, 14/16 /16 month'age.And the in-tervention was accordingly determined as 6 /6 /8 months.After anesthetization, the rats were proceeded with HE staining on musculus gastroc -nemius to test the cross sectional area .Western blot was used to test the expressions of AKT , mTOR, p70S6K and p-p70S6K.Results The muscle wet weight /body weight ratio was significantly increased in 14MEA6, 16MEA6 rats.The centralization of cell nuclei and the keratini -zation of muscle fiber were decreased while the cross sectional area was increased in the preventive electroacupuncture groups .The preventive electroacupuncture was first raised by AKT or phosphorylated AKT expression and the activated p 70S6K promoted the skeletal muscle protein synthesis.Conclusions The appropriate month'age of amyotrophy rat for preventive electroacupuncture is 8 months and the intervention du-ration is 6 months.The effect of preventive electroacupuncture for the aging amyotrophy rat is associated with the pathway IGF /AKT and the preventive electroacupuncture is effective for promoting protein synthesis of rat muscle protein .%目的探讨预防性电针对不同月龄大鼠骨骼肌形态和 IGF /AKT 通路下与蛋白合成相关蛋白表达的影响。方法雄性 SD 大鼠30只,随机分为正常对照组、电针组,各组又分为8月正常组(8 MN),14月龄正常组(14 MC)、16月龄正常组(16 MC)、14月龄电针6个月组(14 MEA6)、16月龄电针6个月组(16MEA6)和16月龄电针8个月组(16MEA8)。麻醉后快速取腓肠肌 HE 染色并测量骨骼肌横截面积,Western 印迹测定大鼠腓肠肌中 AKT、mTOR、p70S6K 和 p-p70S6K 相关蛋白表达。结果14MEA6、16MEA6组 SD 大鼠腓肠肌的肌肉湿重/体重比值显著升高;电针预防性干预的14MEA6、16MEA6组细胞核中心化、肌纤维小角化变少,骨骼肌横截面积显著升高;预防性电针首先通过 AKT 或磷酸化 AKT 表达上调,活化了 p70S6K 促进骨骼肌蛋白合成翻译。结论预防性电针干预增龄性大鼠骨骼肌萎缩的较佳月龄为8月龄,较佳干预周期为6个月。预防性电针干预减缓增龄性大鼠骨骼肌萎缩与 IGF-1/AKT 通路有关,并促进了骨骼肌蛋白合成。
