Objective To study the effects of N-myc downstream regulated gene (NDRG)3 overexpression on the proliferation and migration of Hela cervical cancer cells.Methods After building NDRG3 expression vector, Lipofectamine 2000 transfection reagent was used to establish a stable transfection NDRG3 Hela cervical cancer cell lines, cell proliferation and migration ability were detected using CCK8 and Transwell methods,enzyme-linked immunosorbent assay was utilized to test the expression of CXC chemokine ligand (CXCL)5. Results Overexpression of NDRG3 significantly promoted the proliferation and migration of Hela, and up-regulated the secretion level of CXCL5 of Hela.Conclusions NDRG3 can participate in the cervical cancer process by up-regulating tumor-associated gene CXCL5 level.%目的 研究过表达N-Myc下游调节基因(NDRG)3对Hela宫颈癌细胞增殖和迁移的作用.方法 构建NDRG3过表达载体,利用Lipofectamine 2000转染试剂建立稳定转染NDRG3的Hela宫颈癌细胞株,细胞计数试剂(CCK)8和Transwell方法研究细胞增殖和迁移能力,酶联免疫吸附实验研究趋化因子CXC配体(L)5的表达情况.结果 过表达NDRG3可明显促进Hela的增殖和迁移能力,并上调Hela的分泌性CXCL5蛋白水平.结论 NDRG3可经上调肿瘤相关基因CXCL5的水平参与宫颈癌进程.
