Objective To study the effect of adenovirus-mediated N-ethylamaleimide sensitive fac-tor (NSF) siRNA on cardiac function in rats with myocardial infarction (MI) .Methods Thirty-six adult SD rats were randomly divided into experimental group ,control group and normal saline group (12 in each group) .Two weeks after NSF siRNA recombinant adenovirus vector was injec-ted into the infarcted part on the left ventricular wall ,the LVEF was tested by noninvasive ultra-sonic cardiography ,the LVEDP and +dp/dtmax were measured .The rat heart was removed and cut into serial sections which were stained with TTC .The MI size was observed .Results Two weeks after the NSF siRNA recombinant adenovirus vector was injected ,the LVEF was significantly higher while the LVEDP was significantly lower while the +dp/dtmax was significantly higher in experimental group than in control group and normal saline group ( P< 0 .05 ) .No significant difference was observed in MI size although it was smaller in experimental group than in control group and normal saline group ( P> 0 .05) .Conclusion Injection of adenovirus-mediated NSF siRNA expression vector into the MI can significantly improve the cardiac function in rats with acute MI .However ,it shows no effect on the MI size .%目的：探索腺病毒介导的N-乙基顺丁烯二酰亚胺敏感因子小干扰RNA （NSF-siRNA ）对急性心肌梗死大鼠模型心功能的影响。方法36只成年SD大鼠，随机分为实验组、对照组和生理盐水组，每组12只。实验组经结扎左冠状动脉前降支建立急性心肌梗死模型，模型成功后，大鼠心脏左心室壁梗死区周围局部注射NSF-siRNA重组腺病毒载体。2周后，通过无创超声心动图测定心脏LVEF；用BL-420生物功能实验系统测定左心室舒张末压（LVEDP）及左心室压力最大上升速率（＋dp/dtmax ）；随后取心脏，连续切片行 TTC染色，沿分界线剪下梗死区，测量心肌梗死区质量占全部心脏组织质量的比例，观察心肌梗死范围。结果2周时，实验组LVEF较对照组及生理盐水组明显增加[（46．0±7．5）％ vs （34．0±9．0）％和（27．5±4．5）％，P＜0．05]；LVEDP较对照组及生理盐水组显著降低[（18．5±5．9）mm Hg vs （39．5±24．0）mm Hg和（26．6±24．0）mm Hg （1 mm Hg＝0．133 kPa），P＜0.05]；＋dp/dtmax较对照组及生理盐水组明显增加[（9．7±1．2）mm Hg/s ×103 vs （4．3±2．2）mm Hg/s ×103和（5．2±2．5）mm Hg/s ×103，P＜0．05]；与生理盐水组和对照组比较，实验组心肌梗死范围虽降低，但差异无统计学意义（P＞0．05）。结论心肌梗死周围注射NSF-siRNA腺病毒表达载体，能显著改善大鼠急性心肌梗死后2周心功能，但对心肌梗死范围无明显影响。
