热性惊厥患儿血清中DNA甲基转移酶的表达及其临床意义

摘要

目的 分析热性惊厥患儿血清中DNA甲基转移酶(DNA methyltransferase,DNMT)的表达及其活性变化,探讨其在小儿热性惊厥治疗中的临床意义.方法 选取本院50例热性惊厥患儿纳入热性惊厥组,将本院同期35例健康体检儿童纳入正常对照组,采用酶联免疫吸附测定和荧光定量聚合酶链反应检测两组研究对象血清中DNMT蛋白含量和基因表达水平.结果 热性惊厥组患儿DNMT1、DNMT3A、DNMT3B基因的表达水平均明显高于正常对照组(P<0.001).热性惊厥组患儿血清中DNMT1、DNMT3A蛋白含量均高于正常对照组(P<0.01),但DNMT3B蛋白含量两组间无显著差异(P>0.05);DNMT活性显著高于正常对照组(P<0.01).结论 DNMT蛋白、基因及活性的变化可能参与小儿热性惊厥的发病,抑制表观遗传基因表达可能成为治疗小儿热性惊厥的重要方法.%Objective To explore the change and clinical significance of DNA methyltransferase (DNMT) in children with febrile convulsion. Method 50 children with febrile convulsion in our hospital were enrolled in febrile convulsion group, 35 healthy children during the same period were enrolled in normal control group. Enzyme-linked immunosorbent assay and real-time polymerase chain reaction were used to detect the content of DNMT protein and gene expression in two groups. Result The expression levels of DNMT1,DNMT3A and DNMT3B in febrile convulsion group were significantly higher than those in normal control group (P<0.001). The serum levels of DNMT1 and DNMT3A of febrile convulsion group were higher than those in normal control group (P < 0.01), but there was no significant difference in DNMT3B between the two groups (P > 0.05), the activity of DNMT was significantly higher than that of normal control group (P < 0.01). Conclusion The changes of DNMT protein, gene and activity may be involved in the pathogenesis of febrile convulsion in children, inhibition of epigenetic gene expression may be an important method in the treatment of febrile convulsions in children.
