目的:探讨米非司酮对体外培养的大鼠卵巢颗粒细胞凋亡的影响。方法体外培养SD雌性大鼠的卵巢颗粒细胞,采用流式细胞术及荧光染色观察并分别了解加入米非司酮后细胞的凋亡情况。结果颗粒细胞凋亡数量随米非司酮剂量增加而增多,凋亡率在米非司酮低剂量组与对照组间比较差异无统计学意义(P>0.05),中剂量组、高剂量组高于对照组(P<0.05),各处理组之间比较差异有统计学意义(P<0.05)。结论米非司酮可诱导颗粒细胞发生凋亡,离体颗粒细胞凋亡率随米非司酮剂量增加而增高,可能是米非司酮抑制卵泡发育的机制之一。%Objective To explore the effect of Mifepristone on apoptosis in cultured granulosa cells .Methods The ovarian granulosa cells of SD female rats were cultured in vitro , flow cytometry and fluorometry were conducted to observe the apoptosis of granulosa cells exposed to different concentrations of mifepristone .Results The apoptosis of granulosa cells boosted along with the dose increase of Mifepristone, the apoptosis rate had no significant difference between low -dose group and control group(P>0.05), the apoptosis rate of moderate -dose group and high -dose group were higher than control group ( P<0.05 ) .The results of each treated group had significant differences (P<0.05).Conclusion Mifepristone is able to induce apoptosis in cultured granulosa cells , the apoptosis rate of granulosa cells may be dose -dependent with Mifepristone , which maybe one of the mechanisms of inhibiting ovulation.
