目的 观察单核细胞趋化因子(MCP-1)、基质金属蛋白酶-9(MMP-9)、肿瘤坏死因子(TNF-α)和胰岛素样生长因子(IGF-1)在人主动脉粥样硬化组织中表达的情况.方法 病变组主动脉组织取自外科手术腹主动脉瘤患者,对照组主动脉组织取自非正常死亡健康成人尸体的正常主动脉.用免疫组化方法检测MCP-1,MMP-9,TNF-α和IGF-1在动脉粥样硬化斑块处的表达及定位.结果 病变组MCP-1、TNF-α、MMP-9、IGF-1的表达水平显著高于对照组,差异均有统计学意义(P均<0.05).MCP-1蛋白表达与MMP-9(rs=0.464,P<0.05)、TNF-α表达呈正相关(rs=0.516,P<0.05),但与IGF-1表达无相关性(rs=0.370,P>0.05).结论 MCP-1、MMP-9、TNF-α和IGF-1在人动脉粥样硬化中高表达,MCP-1对MMP-9、TNF-α有促进的作用.%Objective To observe the expressions of monocyte chemoattractant protein-1 ( MCP-1 ), matrix metalloproteinase-9 ( MMP-9 ), tumor necrosis factor-α ( TNF-α ) and insulin-like growth factor-1 ( IGF-1 ) in human atherosclerotic tissue. Methods The atherosclerotic samples of disease group were colleted from the patients with abdominal aneurysm after surgery,and normal aorta samples of control group were collected from health adults died from accidents. The expressions and distributions of MCP-1, MMP-9, TNF-α and IGF-1 in atherosclerotic plaques were detected by using immunohistochemistry technique. Results The expressions of MCP-1, MMP-9 ,TNF-α and IGF-1 were significantly higher in disease group than those in control group ( all P <0.05 ). The expression of MCP-1 was positively correlated to MMP-9 ( rs =0.464,P <0. 05 ) and TNF-α ( rs =0. 516,P <0.05 ),but was not correlated to IGF-1 ( rs = 0.370,P>0.05). Conclusion The expressions of MCP-1,MMP-9,TNF-α and IGF-1 are higher in human atherosclerotic tissue,and MCP-1 has an improvement effect on MMP-9 and TNF-α .
