目的 探讨硫酸镁(MgSO4)对心肺复苏(CPR)后大鼠大脑皮质水肿和水通道蛋白4(AQP4)基因表达的影响.方法 68只健康成年雄性SD大鼠随机分为三组:假手术组、CPR模型组、MgSO4治疗组.每组再分四个亚组:即自主循环恢复(ROSC)后3、6、12、24h组,除了假手术组各时相点为5只大鼠外,另两组大鼠各时相点均为6只.MgSO4治疗组在ROSC后立即行MgSO4干预.应用干湿质量法测定脑皮质含水量(cerebral cortex water content,CCWC),逆转录-聚合酶链反应(RT-PCR)技术测定脑皮质AQP4mRNA的表达水平.结果 MgSO4治疗组ROSC后3、6、12、24 h CCWC和AQP4mRNA表达水平与相同时点CPR模型组比较均明显降低(P<0.05).结论 MgSO4可减轻CPR后脑水肿,其机制可能是降低了AQP4的表达.%Objective To explore the effects of magnesium sulfate(MgSO4)on cerebral edema and expression of aquaporin -4( AQP4) in cerebral cortex after cardiopulmonary resuscitation(CPR) in rats. Methods 68 healthy adult SD rats, were randomly divided into 3 groups; sham(n =20) , CPR model group, MgSO4 treatment group. Each group then divided into 4 subgroups, that is 3 h, 6 h, 12 h, 24 h subgroup after Restoration of Spontaneous Circulation ( ROSC) , each subgroup n = 6, except for sham. The rats of MgSO4 group were treated by MgSO4 at once after ROSC. The cerebral cortex water content(CCWC) was determined by wet - to - dry weight ratio, the level of AQP4 mRNA expression was determined by Reverse Transcription - Polymerase Chain Reaction ( RT - PCR). Results the CCWC and expression level of AQP4 mRNA were Significantly lower in MgSO4 treatment group than the CPR model group (P < 0. 05). Conclusion MgSO4 can reduce cerebral edema after CPR, and its mechanism perhaps down - regulates the expression of AQP4 in brain tissue.
