AIM: To observe the expression and distribution of MMP-2 in the periodontal tissue of first maxillary molar of rat during tooth movement by the orthodontic force. METHODS: 30 SD male rats were grouped according to tooth movement days of 0,1,3,6 and 10d. The orthodontic appliance was applied to set up the model of tooth movement. In situ hybridizasion was used to study the expression of the MMP-2 gene in the periodontium of the first molar . RESULT: In tooth movement groups,the MMP-2 gene expression was observed in osteoclasts, osteoblasts and fibroblasts of periodontium on both sides of movement tooth. The osteoclasts in pressed zone and osteoblasts in tensed zone was more positive stained than that of the controls.CONCLUSION: MMP-2 participated in the remolding of alveolar bone during tooth movement by the orthodontic force.%目的:观察大鼠牙齿正畸移动过程中牙周组织中基质金属蛋白酶-2(MMP-2)在受力下的表达和分布。方法:选取雄性SD大鼠30只,分为空白对照组、牙齿移动1、3、6和10d组。用原位杂交染色方法观察并比较各组中第一磨牙牙周组织中MMP-2的表达和分布。结果:MMP-2基因在受力牙齿压力侧牙周组织的破骨细胞和张力侧牙周组织的成骨细胞和成纤维细胞中的阳性表达明显强于对照组,且随受力时间的增加而增强。结论:本实验证明MMP-2参与了正畸牙齿移动过程中牙周组织的改建。
