不同应力对成骨细胞和细胞骨架影响的实验研究

             

摘要

目的:研究两种不同应力对体外培养的成骨细胞及其细胞骨架形态的影响。方法:分别给第3代的大鼠成骨细胞施加剪切力和静压力,用相差显微 镜和考马斯 亮蓝染色方法观察加力后 不同时间细胞及其细胞骨架形态的变化特征。结果:剪切力作用后6 h,部分细胞出现形态学 、细胞骨架排列等方面的变化;12h后,部分细胞死亡、脱落。较大的静压力(>20kPa)作 用12h后,可引起细胞间隙增大等变化,细胞骨架无明显改变。结论:一定的外力可引起成骨细胞骨架的重排及细胞形态的变化。静压力作为一种类似于生理状 态的外力与剪切力相比,对成骨细胞的形态及细胞骨架影响较轻。%AIM: To examine the effect of two differen t kinds of stress on osteoblast and its cytoskeleton in vitro. METHODS: Fluid shear st ress and compressive pressu re were applied on the rat osteoblasts of the passage third respectively. Phase cont rast microscope and Coomassie BB staining were used to observe the characterist ics of cellular shape and cytoskeleton at different intervals. RESULT S : Changes of the cellular shape, the rearrangment of cytoskeleton wer e found after 6 hours of being applied by shear stress. Some dead cells were also observed after 12 hours. While under the compressive pressure (>20kPa) for 12 h ours, the distance between cells increased and no obvious change of cytoskeleton were observed. CONCLUSION: Stress may cause the rearrange ment of cytoskeleton and the change of cellular shape. The compressive pressure, in comparing to the sheas stress, the later, as a kind of physiological like s tress, has less effect on the cellular shape and cytoskeleton of osteoblasts.
