AIM: The effects of nerve growth factor on the proliferation andcell cycle of human dental papilla mesenchyme cells were detected to evaluate its possible roles in tooth development. METHODS: 3H-TdR incorporation and flow cytometer analysis. RESULTS: The nerve growth factor showed a significant concentration-dependant stimulation of [3H] thymidine uptake, with a maximum increase at 100U/ml, but inhibit the cellular proliferation at 1000U/ml. Nerve growth factor at 100U/ml increased the percenage of cells in S cycle phase and the percentage of S+G2M. CONCLUSION: nerve growth factor stimulated DNA synthesis of human dental papilla mesenchyme cells, and stimulated the transformation of cells from G1 to S cycle phase.%目的:观察神经生长因子对人牙乳头间充质细胞增殖和细胞周期的影响,探讨其在牙胚生长发育过程中的可能作用。方法:采用3H-TdR掺入和流式细胞仪分析法。结果:NGF(1~100U/ml)可明显促进培养的人牙乳头间充质细胞的3H-TdR掺入率。NGF(100U/ml)刺激细胞增殖作用最强。NGF(1000U/ml)抑制细胞的增殖。在NGF(100U/ml)作用下,细胞S%明显升高,细胞增殖指数(S+G2M)%也明显增高。结论:神经生长因子可促进人牙乳头间充质细胞DNA合成,刺激G1期细胞向S期转变。
