2型糖尿病大鼠不同组织中p85mRNA及蛋白表达变化与中药干预作用

摘要

Objective: to investigate of the Variation of Gene Expression of p85 of Type 2 Diabetes Mellitus Rats in Different Tissue and Intervention of Fufang Huanglian Jiangtangpian(abbr:FFHLJTP) on these indexs. Methods: FFHLJTP was given to type 2 diabetic rats. Blood glucose was measured. Blood insulin was measured. The expression of p85mRNA and protein in liver, muscle and fat of type 2 diabetic rats ware determined by RT-PCR and western blot analysis. Results: FFHLJTP treatment decreased the density of blood glucose and blood insulin in type 2 diabetic rats. The mRNA and protein expressions of p85 in liver, muscle and fat were obviously increased. Conclusion: FFHLJTP increases p85mRNA and protein expression in liver, muscle and fat of type 2 diabetic rats. Its mechanism may be partly by raising p85 gene expression in liver, muscle and fat.%目的:观察2型糖尿病大鼠不同组织中p85基因表达的变化及复方黄连降糖片时其变化的影响.方法:对键服佐菌素注射加高脂饮食诱导的2型糖尿病模型大鼠给予复方黄连降糖片治疗8周.测定血糖、血清胰岛素,大鼠肝脏、骨骼肌和脂肪组织p85mRNA的表达及其蛋白的表达.结果:复方黄连降糖片治疗组大鼠的血糖和血清胰岛素均有所下降,各组织够5的基因表达均显著提高.结论:不同组织p85的基因表达的提高可能是复方黄连降糖片治疗2型糖尿病的分子机理之一.
