目的:观察熟地寄生壮骨方对大鼠创伤性骨关节炎模型的影响,探讨其抗炎及保护软骨作用.方法:采用创伤性膝骨关节炎大鼠模型,切取病变膝关节制备膝关节匀浆液,测定给药前后一氧化氮(N0)、肿瘤坏死因子-a(TNF-a)的含量;病变膝关节组织病理切片,按Mankin骨关节炎组织分类法进行评分.结果:与空白组比较,模型组大鼠膝关节NO、TNF-α含量显著增大,大鼠膝骨关节Mankin评分值显著增大;与模型组比较,熟地寄生壮骨方各组NO、TNF-α含量均明显减小,大鼠膝骨关节Mankin评分值均明显减小.结论:熟地寄生壮骨方能降低创伤性骨关节炎模型大鼠膝关节NO、TNF-α含量及改善其软骨形态,达到控制炎症及保护软骨的目的.%Objective:To observe the anti-inflammatory effect and the morphology change of articular cartilage under intervention of Shudi Jisheng Zhuanggu decoction (SJZ) in knee post-traumatic osteoarthritis model rats.Methods:Rats were removed the medial collateral ligament and the medial meniscus to build models of knee post-traumatic osteoarthritis (Hulth's modeling method)before administration of drugs.Prepared 20% homogenization buffer of the knees to measure the contents of nitric oxide (NO) and TNF-α.Histologic sections of cartilage were received HE staining and Gemini staining were evaluated according to Mankin cartilage histological score.Results:Mankin score and the contents of NO and TNF-α were significantly increased in model group compared to control group.Mankin score and contents of NO and TNF-α of SJZ groups were significantly decreased compared to model group.Conclusion:SJZ can control inflammation and ameliorate morphology of articular cartilage in rats with knee Post-traumatic osteoarthritis,so as to protect the cartilage and delay cartilage degeneration.
