糖尿病大血管病变“气阴两虚”态模型的建立

摘要

目的:建立糖尿病大血管病变“气阴两虚”态模型.方法:60只雄性GK大鼠(随机血糖＞11.1 mmol.L-1),按随机数字表法分为GK组、模型组、中药组各20只,另设正常Wistar大鼠20只为正常组.正常组、GK组喂饲普通饲料,模型组和中药组喂饲高脂饲料,造模同时中药组给予益气养阴中药治疗.5周后检测各组大鼠糖脂代谢、胰岛素抵抗指数(HOMA-IR)、血管内皮细胞凋亡和Caspase-3表达等指标,光镜下观测大血管组织病理.结果:与正常组和GK组比较,模型组大鼠血糖、TC和TG、胰岛素抵抗、大血管内皮细胞凋亡和腹主动脉病理比较差异有统计学意义,与模型组比较中药组所有指标均有显著改善.结论:通过GK大鼠喂养高脂饲料的方法可制备糖尿病大血管病变“气阴两虚”态动物模型.%Objective:To establish models of Qi and Yin deficiency state and syndrome in diabetes mellitus macroangiopathy.Methods:20 Wistar rats were included in the control group,while 60 male GK rats (random blood glucose ＞ 11.1 mmol/L)were randomly divided into GK group,model group and Chinese traditional medicine (TCM)group,20 rats in each group.Control group and GK group feed ordinary,model group and TCM group eat with high fat feed.TCM group had a Chinese medicine treatment at the same time.After 5 weeks,testing fasting blood ghcose、fasting serum insulin (FINS),insulin resistance index (HOMA IR),Total cholesterol;triglycerides and vascular endothelial cell apoptosis;caspase-3 express;Light microscope observe pathology of the Abdominal aorta.Results:compared with the control group and GK group,the blood glucose,TC,TG,insulin resistance,vascular endothelial cell apoptosis,and large blood vessels tissue pathological have significant differences.Compared with the control group,TCM group all the indexes have improved significantly.Conclusion:By feeding the GK rats with high-fat feed,models of Qi and Yin deficiency state and syndrome in type 2 diabetes mellitus macroangiopathy was successfully established.
