禽内源性反转录病毒ALVE1在鸡基因组上下游序列鉴定

摘要

已知鸡基因组中含有禽内源性反转录病毒ALVE1序列,而ALVE1在鸡基因组中的上游和下游区域位置在参考基因组中并未成功鉴定.本研究成功鉴定出ALVE1所在鸡基因组中的上、下游序列,并通过提取CEF细胞和DF-1细胞基因组反过来验证了ALVE1的位置信息.与参考基因组不同的是,我们发现ALVE1序列包含3'LTR序列,在鸡1号染色体上的插入位置为chr1:65,995,534~65,993,540.本研究不仅提供了ALVE1在鸡基因组中的完整信息,也为进一步研究ALVE1的功能奠定了基础.%The chicken genome contains endogenous avian leukosis viral element on chromosome 1 (ALVE1). However, the upstream and downstream sequences flanking region of the ALVE1 element remains unidentified in the reference genome. In this study, the upstream and downstream sequences of ALVE1 in the chicken genome were identified using RACE and inverse PCR, and confirmed by PCR amplification. In addition, the location of ALVE1 was verified using CEF and DF-1 cell genomes. Here, the complete sequences containing the upstream and downstream of ALVE1 element were figured out with genomic DNA from CEF cells. Unlike the reference genome, ALVE1 element contained 3'LTR sequences on chicken chromosome 1 (position: 66086818-66086750). This study not only provided the complete information of ALVE1 in the chicken genome but also provided a basis for further study on the function of ALVE1.
