正中隆起室管膜细胞在高血压大鼠的形态学改变

     

摘要

目的:观察大鼠正中隆起室管膜细胞在高血压不同时期的形态学变化,探讨该室周器官在高血压发病过程的作用.方法:选择健康雄性Wistar大鼠60只,随机分为对照组,左旋-硝基精氨酸1、2、3、4周组.制备高血压动物模型,检测巢蛋白的表达情况.结果:用药动物组血压明显高于对照组,模型制备成功.光镜下观察到高血压初期(1、2周组)正中隆起室管膜细胞胞体增大,室管膜下细胞排列密集,室管膜下血管扩张.高血压后期(3、4周组)室管膜细胞排列稀疏,室管膜下细胞深染,软膜下血管扩张明显.巢蛋白免疫阳性细胞在高血压初期多出现在正中隆起室管膜下层,在高血压后期多出现在室管膜层.结论:正中隆起室管膜细胞与高血压发生有密切联系.%Objective:To observe the morphological changes of the ependymal cells in the median eminence of hypertensive rats at different time,and to infer the role of median eminence in the process of hypertension.Methods:60 healthy male Wistar rats with body weight between 200-250 g,were randomly divided into five groups:a control group treated with physiological saline (n=12),one-week treatment group treated with L-NNA for one week,two-week treatment group treated with L-NNA for two weeks,three-week treatment group treated with L-NNA for three weeks,four-week treatment group treated with LNNA for four weeks.The process was as follows:preparation for the animal model of hypertensive rats; preparation of light microscopic specimen; detection of the expressions of nestin; and statistical analysis.Results:The blood pressure of the treatment groups was apparently higher than that of the control group.The animal model of hypertensive rats was successfully established and the morphological changes on the ependymal cells in the median eminence were successfully observed.In the early days of hypertension (one-and two-week group),the volume of ependymal cells in the median eminence greatened and spread closely,and the blood vessels below ependyma extended.In the later period (three-and four-week group),the ependymal cells spread sparsely,the cells below ependyma were coloured deeply,and the blood vessels below soft mask extended apparently.The nestin immuno-positive cells were distributed mostly in the sub-layer of ependyma in the early days of hypertension,but distributed mostly in the layer of ependyma in the later period.Conclusion:The ependymal cells in the median eminence has a close relationship with the process of hypertension.
