小鼠肾发育中Ki67的表达

         

摘要

目的:探讨小鼠肾发育中细胞增殖的变化规律.方法:应用免疫组织化学与免疫印迹,并结合体视学分析的方法,对小鼠肾发育中的细胞增殖进行定性和定量观察.结果:免疫组织化学结果显示生肾区Ki67的阳性细胞在肾发育的早期高度密集,P7 d时消失;髓放线和外髓Ki67的阳性细胞在P7 d之后,仍保持着较高的增殖率;免疫印迹结果显示皮质区Ki67的蛋白表达从E16 d~P1 d逐渐升高,以后则逐渐降低,而髓质区Ki67的蛋白表达在P3 d达到高峰.结论:小鼠肾发育中肾皮质比肾髓质更早达到成熟状态,髓质在很长时间内是肾最不成熟的结构.%Objective: To explore the change of cell proliferation during mouse kidney development. Methods: Different developmental mouse kidneys were used to investigate the cell proliferation with immunohistochemistry technique, Western blotting and stereological methods. Results: During the prophase of mouse kidney development, the expression of Ki67 was mainly concentrated in metanephrogenic blastema influence, and disappeared at P7 d. The positive cells of Ki67 were still to remain higher growth rate in medullary ray and medulla external after P7 d. The expression of Ki67 protein of renal cortex gradually heightened from E16-P1 d, and reduced after PI d. However, the expression of Ki67 protein of renal medulla reached peak at P3 d. Conclusion: During mouse kidney development, the renal cortex became mature before renal medulla, and renal medulla was the most immature structure during long phase.
