Bcl-2和Hrk及细胞周期素E在胃癌组织中的表达及其意义

摘要

目的 探讨凋亡抑制蛋白Bcl-2、凋亡促进蛋白Hrk及细胞周期素E(Cyclin E)在胃癌中的表达及其意义.方法 采用免疫组化(S-P)法检测58例胃癌患者的癌组织蜡块标本及正常癌旁组织中Bcl-2蛋白、Hrk蛋白及Cyclin E蛋白的表达情况.结果 Bcl-2蛋白在胃癌组织中阳性表达38例,阳性率为65.5%,癌旁组织中阳性表达14例,阳性率为24.1%,两种组织中Bcl-2蛋白阳性率比较,差异有统计学意义(χ2=20.077,P<0.05).不同肿瘤分化程度、临床分期及有无淋巴转移胃癌患者中Bcl-2蛋白阳性率比较,差异均有统计学意义(P<0.05);不同年龄、肿瘤直径、Borrmann分型、浸润深度胃癌患者中Bcl-2蛋白阳性率比较,差异均无统计学意义(P>0.05).Hrk蛋白在胃癌组织中阳性表达12例,阳性率为20.7%,癌旁组织中阳性表达36例,阳性率为62.1%,两种组织中Hrk蛋白阳性率比较,差异有统计学意义(χ2=20.471,P<0.05).Cyclin E蛋白在胃癌组织中阳性表达34例,阳性率为58.6%,癌旁组织中阳性表达11例,阳性率为18.9%,两种组织中Cyclin E蛋白阳性率比较,差异有统计学意义(χ2=19.206,P<0.05).Spearman相关分析表明,Bcl-2蛋白与Hrk蛋白呈负相关(r=-0.286,P<0.05),与Cyclin E蛋白呈正相关(r=0.749,P<0.05).结论 胃癌的形成与多种蛋白的相互作用及基因水平上正负信号转导、调控密切相关,凋亡抑制蛋白Bcl-2、凋亡促进蛋白Hrk及Cyclin E共同参与胃癌的发生发展.%Objective To study the expression and significance of inhibitor of apoptosis protein Bel - 2, apoptosis promote protein Hrk and cell cycle element Cyclin E in gastric cancer. Methods The expressions of Bel - 2, Hrk and Cyclin E in the samples of gastric cancer and adjacent normal tissues of 58 patients with gastric cancer were detected by S - P method. Results Bel -2 protein had positive expression in 38 cases of gastric cancer tissues with a positive rate of 65. 5% , and had positive expression in 14 cases of adjacent normal tissues with a positive rate of 24. 1 %. The difference of positive rate between the two tissues showed statistically significant difference ( x2 = 20. 077, P < 0. 05 ) . Patients with different cancer differentiation levels , clinical stages and whether having lymph node metastases or not showed statistically significant difference in the positive rate of Bel -2 protein ( P <0. 05 ) . Patients with different age, tumor diameter, Borrmann type and infiltration depth showed no statistically significant difference in the positive rate of Bel -2 protein ( P >0. 05 ) . Hrk protein had positive expression in 12 cases of gastric cancer tissues with a positive rate of 20. 7% , and had positive expression in 36 cases of adjacent normal tissues with a positive rate of 62. 1 %. The difference of positive rate between the two tissues showed statistically significant difference ( x2= 20. 471 , P < 0. 05 ) . Cyclin E protein had positive expression in 34 cases of gastric cancer tissues with a positive rate of 58. 6% , and had positive expression in 11 cases of adjacent normal tissues with a positive rate of 18.9%. The difference of positive rate between the two tissues showed statistically significant difference ( x2 = 19. 206, P <0. 05 ) . Spearman correlation analysis showed that Bcl - 2 protein was negatively correlated with Hrk protein ( r = - 0. 286 , P < 0. 05 ), and was positively correlated with Cyclin E protein ( r =0. 749, P <0. 05 ) . Conclusion The pathogenesis of gastric cancer is closely related to the mutual effect of various proteins and positive and negative signal transduction and regulation in genetic level. Inhibitor of apoptosis protein Bcl - 2, apoptosis promote protein Hrk and Cyclin E are involved in the pathogenesis and development of gastric cancer.
