复方脑康胶囊对β-淀粉样肽诱导SH-SY5Y细胞凋亡的保护作用研究

         

摘要

OBJECTIVE: To study the protective effect of Compound naokang capsules on human neuroblastoma cell line SH-SY5Y cell apoptosis induced by β-peptide25-35(Aβ25-35). METHODS: SH-SY5Y cells were divided into 3 groups after inoculation: control group and (ApVss (25 utnol-L"1) group and Compound naokang capsule (0.725 g.mL-1)+Aβ25-35(25 umol.L-1)- Cell survival rate(MTT), LDH leakage rate, axonal length, area of cell body were determined, and neroprotective effect of Compound naokang capsules on Aβ25-35 was observed. RESULTS: Compared with control group, axonal length and area of cell body of SY5Y cells in damaged group were small, MTT metabolic rate reduced and LDH leakage rate increased. Above parameters turned into normal level after adding Compound naokang capsules aqueous extract. CONCLUTION: Compound naokang capsules have neuropro-tective effect. It can diminish neurotoxicity of Aβ25-35.%目的:研究复方脑康胶囊对β-淀粉样肽(Aβ)25-35(Aβ25-35)诱导人神经母细胞瘤SH-SY5Y细胞凋亡的保护作用.方法:将SH-SY5Y细胞接种后分为对照、Aβ25-35(25μmol·L-1)和复方脑康胶囊(0.725 g·mL-1) +Aβ25-35(25 μmol· L-1)组.通过测定细胞存活率(MTT法)、乳酸脱氢酶(LDH)漏出率、细胞轴突长度、胞体面积,观察复方脑康胶囊对Aβ导致神经毒性的保护作用.结果:与对照组比较,Aβ25-35组SY5Y细胞的轴突长度和胞体面积均较小,MTT代谢率降低,LDH漏出率升高,而加入复方脑康胶囊水提液后,可使上述指标恢复或接近正常.结论:复方脑康胶囊具有神经保护作用,可减轻AB导致的神经毒性.
