NSCLC患者血清miR系列基因表达水平及与化疗疗效的相关性研究

         

摘要

目的 探讨非小细胞肺癌(non-small lung cancer, NSCLC)患者血清miR (microRNA)系列基因表达水平及与化疗疗效的相关性.方法 2011年3月~2016年9月选择200例未经任何抗癌治疗的NSCLC患者作为NSCLC组,100例健康志愿者作为健康组,分别收集血清样本;25例晚期NSCLC患者化疗前和化疗后的血清样本.采用qRT-PCR方法检测NSCLC组和健康组中血清miR系列基因表达水平,判断血清miR系列基因表达水平与预后的关系.结果 NSCLC组的血清miR-19a、miR-19b-l、miR-20a、miR-17-3p、miR-18a 和miR-92-1基因表达与健康组存在差异(P<0.01).NSCLC患者的血清上述基因表达化疗前与化疗后存在明显差异(P<0.05);健康组的血清miR-19a、miR-19b-l、miR-20a、miR-17-3p、miR-18a 和miR-92-1基因表达与化疗前患者存在明显差异(P<0.05), 与化疗后的患者比较,差异无统计学意义(P>0.05).结论 NSCLC患者血清存在高水平miR系列基因表达,化疗可降低miR系列基因表达水平,上述6种miR基因血清表达有助于NSCLC的诊断、预测和预后的判断,联合检测有助于敏感性的提高.%Objective To investigate the correlation between the expression levels of serum miR(microRNA) gene and chemotherapy efficacy in the patients with non-small cell lung cancer(NSCLC). Methods From March 2011 to September 2016, 200 NSCLCpatients without any anticancer treatment were selected as the NSCLCgroup. 100 healthy volunteers were selected as the healthy group. Serum samples were collected separately; serum samples before and after chemotherapy in 25 patients with advanced NSCLCwere collected. The qRT-PCRmethod was used to detect the expression level of serum miRgene in NSCLCgroup and healthy group, and the relationship between the expression level of serum miRgene and prognosis was determined. Results The serum levels of miR-19a, miR-19b-l, miR-20a, miR17-3p, miR-18a, and miR-92-1 in the NSCLCgroup were different from those in the healthy group(P<0. 01). There were significant differences in the expressions of the above genes before and after chemotherapy in the serum of NSCLCpatients (P<0. 05); the serum levels of miR-19a, miR-19b-l, miR-20a, miR-17-3p, miR-18a and miR-92-1 in the healthy group were significantly different from those before chemotherapy (P<0. 05). There was no statistical difference compared with the patients after chemotherapy (P>0. 05). Conclusion There is a high level of miRgene expression in the serum of NSCLCpatients. Chemotherapy reduces miRgene expression levels. The above six miRgene serum expressions contribute to the diagnosis, prediction and prognosis of NSCLC. The joint detection helps increase sensitivity.
