溃疡性结肠炎模型小鼠调节性T细胞特点的研究

摘要

目的 研究溃疡性结肠炎(UC)模型小鼠调节性T细胞(Treg)的特点.方法 制备葡聚糖硫酸钠诱导的UC模型小鼠(UC组,n=5),检测UC组小鼠和正常小鼠(对照组,n=5)外周血、肠系膜淋巴结、回肠固有层黏膜、结肠固有层黏膜、脾脏的Treg水平、Treg中叉头框蛋白3(Foxp3) mRNA水平及单个核细胞上清培养液中白细胞介素10(IL-10)水平.结果 外周血、肠系膜淋巴结、回肠固有层黏膜、结肠固有层黏膜中,UC组Treg水平明显高于对照组[(2.288±0.021)%比(1.032 ±0.147)%、(3.856 ±0.684)%比(2.164 ±0.088)%、(2.702±0.316)%比(1.568±0.187)%、(14.240±1.693)%比(4.786±0.470)%],Treg中Foxp3 mRNA水平均低于对照组[(0.438±0.031)比(0.771 ±0.039)、(0.559±0.016)比(0.717±0.034)、(0.349±0.067)比(0.549±0.020)、(0.435±0.086)比(0.746±0.044)],单个核细胞上清培养液中IL-10表达水平低于对照组[(0.430±0.430) ng/L比(3.873±0.587) ng/L、(1.115±0.645) ng/L比(3.340±0.121)ng/L、(2.493±0.157) ng/L比(5.284±0.238) ng/L、(2.253±0.248) ng/L比(4.383±0.077)ng/L],差异均有统计学意义(均P<0.05);2组脾脏中Treg水平、Treg中Foxp3 mRNA水平、单个核细胞上清培养液中IL-10表达水平比较,差异均无统计学意义(均P>0.05).结论 UC的发病与免疫异常有关,UC模型鼠不同组织中Treg数量增多且存在功能缺陷.%Objective To investigate the characteristics of regulatory T (Treg) cells in dextran sulphate sodium (DSS) induced ulcerative colitis (UC) model in mouce.Methods DSS-induced UC model was established in 5 mice (UC group);the Treg cell level, forkhead box protein p3 (Foxp3) mRNA level of Treg cell, secretive interleukin (IL)-10 level of mononuclear cells in peripheral blood (PB), mesenteric lymph node (MLN), lamina propria (LP) of jejunum and colon, spleen (SP) were compared between UC group and normal mice (control group, n =5).Results In multiple samples including PB, MLN, LP of jejunum and LP of colon, the Treg cell levels were significantly higher in UC group than those in control group [(2.288 ± 0.021) % vs (1.032 ± 0.147) %, (3.856 ± 0.684)% vs (2.164±0.088)% ,(2.702 ±0.316)% vs (1.568 ±0.187)% ,(14.240 ± 1.693)% vs (4.786 ± 0, 470)%], the Foxp3 mRNA levels of Treg cell were significantly lower in UC group than those in control group [(0.438±0.031) vs (0.771 ±0.039),(0.559 ±0.016) vs (0.717 ±0.034),(0.349 ±0.067) vs (0.549 ± 0.020), (0.435 ± 0.086) vs (0.746 ± 0.044)], the secretive IL-10 levels of mononuclear cells in UC group were significantly lower than those in control group [(0.430 ± 0.430) ng/L vs (3.873 ± 0.587) ng/L, (1.115 ±0.645) ng/L vs (3.340 ±0.121) ng/L, (2.493 ±0.157) ng/L vs (5.284 ±0.238) ng/L, (2.253 ±0.248) ng/L vs (4.383 ± 0,077) ng/L];the Treg cell level, Foxp3 mRNA level of Treg cell, secretive IL-10 level in SP were not significantly different between groups (P > 0.05).Conclusions The mechanism of UC is related with abnormal immunological function;in UC mice, the number of Treg cell is increased, but associated with functional defect.
