黄蘑多糖对H22荷瘤小鼠肿瘤生长及其VEGF表达的影响

     

摘要

Objective: To observe the anti-tumor effect of huangmo polysaccharides on Hepatoma-22 (H22) tumor-bearing mice, and to explore whether it was related with improving immunity and inhibiting tumor angiogenesis. Methods: The tumor-bearing mice model was established by inoculating H22 cell suspension axillarilly. 24 hours later, all mice were given chemicals by oral administration once a day, for 12 days. Then mice were killed, and the tumors, spleens and thymuses were taken and weighed. The anti-tumor rate, the spleens and thymus index were calculated, blood were taken by eye balls, nucleated cells of blood was also counted soon. The tumor specimens were treated by frozen section immunohisto -chemistry method to detect the level of VEGF expression of tumor tissue. Results: The tumor weight in each huangmo treated group was lower than the model group, which was significant in the high dose group (P<0.05); each immune index of huangmo treated group was significantly higher than the CTX group and model group (P<0.01 or P<0.05); the VEGF expression levels in each huangmo treated group were significantly lower than the model group (P<0.01). Compared with the CTX group, the difference of VEGF expression in medium and low group was significant, there was no statistically significant difference of VEGF expression in high dose group (P>0.05). Conclusion: Humangmo polysaccharides can depress the growth of tumor on H22 tumor-bearing mice in certain extent. It can reduce the expression levels of VEGF in the tumor and also can be indentified as biological response modifiers, which acts by enhancing immune system to achieve anti-tumor effect.%目的:观察黄蘑多糖对H22(小鼠肝癌细胞系)荷瘤小鼠的抑制肿瘤效果,并探讨该机制是否与提高机体免疫力和抑制肿瘤血管生成有关.方法:采用腋下接种癌细胞悬液建立荷瘤小鼠模型,造模成功24 h后灌胃给药,连续给药12 d后,处死动物,剥离瘤块、胸腺、脾脏并分别称重,计算抑瘤率、胸腺指数、脾指数;眼球取血计数有核细胞数;以冰冻切片免疫组织化学法检测各组瘤组织血管内皮生长因子(VEGF)表达程度.结果:黄蘑多糖各组的肿瘤重量均低于模型组,其中,中、高剂量组的抑瘤效果显著(P<0.05),且各项免疫指标显著高于环磷酰胺组(CTX组)和模型组(P<0.01或P<0.05);黄蘑多糖各组的VEGF表达率显著低于模型组(P<0.01),黄蘑多糖低、中剂量组VEGF的表达与CTX组比较,差异有统计学意义(P<0.05),高剂量组与CTX组比较,差异无统计学意义(P>0.05).结论:黄蘑多糖对H22移植瘤的生长有一定抑制作用,可降低瘤组织中VEGF的表达,亦可作为生物反应调节剂,通过增强机体免疫功能等作用达到抗肿瘤的功效.
