Objective: To study the effect of blood loss on plasminogen activator inhibitor-1 (PAI-1) on the coagulation and fibrinolysis system in rats. Methods: The rats were randomly divided into three groups (10 animals in each group), acute blood loss group (group A), chronic blood loss group (group B) and control group (group C). After making animal model of anemia, we detected the content of hemoglobin (HB) in blood. Then, we tested the level of PAI—1 in plasma by ELISA in consecutive 8 days without any intervention. Results: The PAI-1 level increased in plasma when blood lost and HB decreased, and there was significant difference between control group and blood loss group. In chronic blood loss group, the content of PAI-1 in plasma increased markedly at first, and lastly maintained a high level with the increasing in blood loss and decreasing in HB, In acute blood loss group, the level of PAI-1 reached a peak and fell rapidly, then increased slowly, and the range of increase was lower than that of chronic blood loss group. Conclusion: The level of PAI-1 increases in plasma with blood lost increasing and HB decreasing.%目的:观察失血对大鼠凝血纤溶系统中纤溶酶原激活物抑制物-1(plasminogen activator inhibitor-1,PAI-1)的影响.方法:将大鼠随机分为急性失血组(A组)、慢性失血组(B组)和对照组(C组),每组10只.制备动物贫血模型,采血后先检测血红蛋白(HB)并记录,血标本离心后分离血浆,ELISA法检测血浆中PAI-1的含量,共观察8 d,期间不采取任何干预措施.结果:大鼠失血、HB降低后血浆中PAI-1含量增加,其中,A组和B组PAI-1的含量较对照组差异明显,B组中随失血量的增加和HB的降低,血浆中PAI-1的含量显著增加后维持在这一高水平.A组急性大量失血后PAI-1含量先达高峰后迅速下降,随后缓慢增加,但增加的幅度不及B组.结论:大鼠血浆中PAI-1的含量随失血量的增加和HB降低而增高.
